乌梅丸改善肥胖小鼠血管内皮功能的作用机制研究OA
Study on the mechanism of Wumei Pill(乌梅丸)in improving vascular endothelial function of obese mice
目的 探讨乌梅丸(WMP)对肥胖小鼠血管内皮功能的作用及对氧化低密度脂蛋白(ox-LDL)诱导的血管内皮损伤的影响及机制.方法 采用高脂饮食诱导肥胖小鼠模型,共喂养24周,随后将模型建立成功的小鼠随机分为模型组、乌梅丸低、中、高剂量组、二甲双胍组.对照组和模型组给予生理盐水灌胃,其余组给予相应的药物灌胃,共干预7周.记录体质量,采用HE染色观察主动脉形态学变化,检测血清血脂含量,ELISA法检测各组小鼠主动脉炎症因子水平.体外培养HUVECs,应用CCK-8法、TUNEL法、成管实验、划痕实验、Transwell实验分别检测细胞增殖、凋亡、成管、迁移能力.结果 1)与模型组比较,乌梅丸高剂量组体质量、血脂、丙二醛含量下降(P<0.05),过氧化氢酶水平上升(P<0.01),白介素1β、白介素6水平降低(P<0.001),HE染色显示主动脉组织形态得到改善;2)与ox-LDL组比,10%WMP组细胞增殖、迁移能力提升(P<0.05),凋亡能力显著下降(P<0.001).结论 乌梅丸可改善肥胖小鼠的血管内皮功能、减轻ox-LDL诱导的血管内皮细胞损伤,其机制可能是降低炎症反应、抗氧化,促进内皮细胞增殖迁移、抑制细胞凋亡.
Objective To investigate the effects and mechanisms of Wumei Pill(乌梅丸,WMP)on endothelial function in obese mice and its impact on oxidized low-density lipoprotein(ox-LDL)-induced endothelial injury.Methods A high-fat diet(HFD)-induced obe-sity mouse model was established by feeding the mice for 24 weeks.Afterwards,the successfully established obese mice were randomly divided into the following groups:model group,the three WMP-treated groups at low,medium and high doses,and metformin group.The control group and model group were administered normal saline via gavage,while the other groups received the corresponding drug treatments via gavage for an additional 7 weeks.Body weight was recorded,using HE staining to observe morphological changes in the aorta,detecting serum lipid levels,and employing ELISA to measure inflammatory cytokine levels in each group.In vitro,HUVECs were cultured,and the proliferation,apoptosis,tubule formation,migration,and transwell migration abilities of the cells were assessed using CCK-8 assay,TUNEL assay,tubule formation assay,scratch assay,and transwell assay,respectively.Results Compared with the model group,the high-dose WMP group showed significant decrease in body weight,serum lipids,and malondialdehyde levels(P<0.05),while catalase activity was increased(P<0.01).Additionally,the levels of interleukin-1β and interleukin-6 were significantly reduced(P<0.001).HE staining revealed improved morphological changes in the aortic tissue.Compared to the ox-LDL group,the 10%WMP group showed enhanced cell proliferation and migration capacity(P<0.05)and a significant reduction in apoptosis(P<0.001).Conclusion WMP can improve endothelial function in obese mice and alleviate ox-LDL-induced endothelial cell injury,with its mechanism possibly involving reducing inflammation,antioxidant effects,promoting endothelial cell proliferation and migration,and inhibiting cell apoptosis.
