饮食因素与睡眠障碍风险的孟德尔随机化研究:基于大规模GWAS数据的因果推断OA
Mendelian randomization study of dietary factors and sleep disorder risk:causal inference based on large-scale GWAS data
目的:探索不同种类饮食与睡眠障碍间的关系.方法:采用两样本孟德尔随机化(MR)分析欧洲人群38种饮食因素(PMID:35653391)与睡眠障碍数据(430 194例病例)的因果关系.通过逆方差加权法(IVW)、加权中位数法、MR-Egger及加权模式法评估饮食因素与睡眠障碍的潜在因果关系.结果:IVW分析显示,水果摄入量(OR=0.756,95%CI:0.643~0.883,P=0.001)、果干摄入量(OR=0.550,95%CI:0.376~0.803,P=0.002)与睡眠障碍呈负相关.果干摄入量经FDR校正后显著性减弱,且经其他方法(加权中位数法、加权模式法)验证后仍支持上述结论.红肉、羊肉及鱼类等摄入未观察到显著关联.结论:水果摄入、果干摄入与睡眠障碍之间存在负向因果关系,提示可能具有保护作用.本研究为饮食干预睡眠障碍提供了遗传学证据,未来需结合生物学机制进一步验证.
Objective:To investigate the relation between different dietary factors and sleep disorders.Methods:A two-sample Mendelian randomization(MR)analysis was conducted to investigate the causal relationship between 38 dietary factors(PMID:35653391)and sleep disorders(430 194 cases)in European populations.Potential causal associations between dietary factors and sleep disorders were assessed using inverse variance weighted(IVW),weighted median,MR-Egger,and weighted mode methods.Results:IVW analysis showed that fruit intake(OR=0.756,95%CI:0.643-0.883,P=0.001)and dried fruit intake(OR=0.550,95%CI:0.376-0.803,P=0.002;)were negatively associated with sleep disorders.Although the significance of dried fruit intake weakened after FDR correction,the conclusion was still supported by other methods(weighted median,weighted mode).No significant associations were observed for the intake of red meat,lamb,or fish.Conclusions:Fruit intake and dried fruit intake have a negative causal association with sleep disorders,suggesting a potential protective effect.This study provides genetic evidence for dietary interventions in sleep disorders,and future studies are needed to further validate the findings with biological mechanisms.
