目的:探索肉苁蓉总苷(glycosides of cistanche,GCs)对帕金森病(Parkinson's disease,PD)模型小鼠黑质多巴胺(dopamine,DA)能神经元的保护作用及量效关系.方法:将60只C57BL/6雄性小鼠随机分为空白组、模型组、GCs给药组(50、100、200 mg/kg)和阳性组,每组10只.除空白组外其余组均腹腔注射MPTP造模,各剂量GCs组和阳性组给予相应剂量灌胃给药.通过行为学实验评估小鼠的运动功能,免疫组织化学染色法和Western blot法分别检测小鼠中脑黑质致密部酪氨酸羟化酶(tyrosine hydroxylase,TH)阳性细胞数量及蛋白表达量,ELISA法测定小鼠纹状体DA含量.结果:与空白组比较,模型组小鼠运动能力下降(P<0.01),中脑黑质致密部的TH阳性细胞数和TH蛋白表达减少(P<0.01),纹状体DA含量降低(P<0.01);与模型组比较,100 mg/kg GCs组能显著改善上述指标(P<0.05),且效果优于50和200 mg/kg GCs组(P<0.05).结论:GCs对PD模型小鼠DA能神经元具有神经保护作用,并在100 mg/kg剂量下治疗效果较佳.

Objective:To explore the protective effects and dose-effect relationship of glycosides of cistanche(GCs)on dopamine(DA)neurons in the substantia nigra of Parkinson's disease(PD)model mice.Methods:Sixty male C57BL/6 mice were randomly divided into the control group,model group,GCs administration groups(50,100,200 mg/kg)and positive control group,with 10 mice in each group.Except for the control group,all other groups were intraperitoneally injected with MPTP to establish the PD model.The GCs groups and the positive control group were administered corresponding doses by intragastric administration.Behavioral tests were performed to evaluate the motor function of mice.Immunohistochemical staining and Western blot were used to detect the number of tyrosine hydroxylase(TH)-positive neurons and TH protein expression level in the substantia nigra pars compacta of the midbrain,respectively.The DA content in the striatum was determined by ELISA.Results:Compared with the control group,the model group presented decreased motor ability(P<0.01),reduced TH-positive cells and TH protein expression in the substantia nigra pars compacta of the midbrain(P<0.01),and decreased DA content in the striatum(P<0.01).Compared with the model group,the 100 mg/kg GCs group significantly improved the above indicators(P<0.05),and the effect was better than that of the 50 and 200 mg/kg GCs groups(P<0.05).Conclusion:GCs have a neuroprotective effect on DA neurons in PD model mice,with an optimal therapeutic effect at the dose of 100 mg/kg.
