目的:阐明粉尘螨可培养共生细菌的组成,分析其组成特征及多样性水平.方法:采用细菌分离培养方法对实验室饲养的粉尘螨可培养共生细菌进行分离和纯化,通过观察细菌形态特征、革兰氏染色,对分离纯化的菌株进行PCR扩增测序鉴定,并采用系统发育树分析其进化关系;同时,统计各分类水平优势单元的丰度,对多样性水平进行分析.结果:从粉尘螨体内分离到16株菌株,其中革兰阳性菌14株,革兰阴性菌2株;4株细胞形态为短杆状,1株为长杆状,11株为球状.经鉴定,16株细菌分属于3门3纲6目7科9属16种,优势门、纲、目、科、属、种分别为放线菌门(Actinomycetota,84.5%)、放线菌纲(Actinomycetes,84.5%)、微球菌亚目(Micrococcus suborder,83.4%)、微球菌科(Micrococcaceae,83.1%)、考克氏菌属(Kocuria,68.3%)、马赛考克氏菌(Kocuria massiliensis,68.3%).结论:粉尘螨可培养共生细菌具有较高的多样性,系统发育树分析表明这些细菌具有较高的进化多样性,为后续通过基因组学解析菌群功能基因、开发基于内共生菌的新型生物防治方法奠定了基础.

Objective:To clarify the composition of culturable symbiotic bacteria in Dermatophagoides farinae,and analyze their composition characteristics and diversity levels.Methods:Bacterial isolation and culture methods were used to isolate and purify culturable symbiotic bacteria from laboratory-reared Dermatophagoides farinae.The isolated and purified strains were identified through morphological characteristics,Gram staining,and PCR amplification and sequencing,and their evolutionary relationship was analyzed by phylogenetic tree.Additionally,dominant taxa at each classification level were counted and the diversity levels were analyzed.Results:Sixteen strains were isolated from Dermatophagoides farinae,including 14 Gram-positive bacteria and 2 Gram-negative bacteria.The cell morphology of 4 strains was short rod,1 strain was long rod and 11 strains were spherical.The 16 strains were identified as belonging to 3 phyla,3 classes,6 orders,7 families,9 genera and 16 species.The dominant phyla,class,order,family,genus and species were Actinomycetota(84.5%),Actinomycetes(84.5%),Micrococcus suborder(83.4%)and Micrococcaceae(83.1%),Kocuria(68.3%)and Kocuria massiliensis(68.3%),respectively.Conclusions:The culturable symbiotic bacteria of Dermatophagoides farinae have high diversity.Phylogenetic tree analysis indicates considerable evolutionary diversity among these bacteria.This study lays the foundation for subsequent genomic analysis of bacterial functional genes and the development of novel biocontrol strategies based on endosymbiotic bacteria.
