柴芍解郁颗粒质量标准研究OA
Study on the quality standard for Chaishao Jieyu granules
目的 建立柴芍解郁颗粒的质量标准.方法 采用薄层色谱法(TLC)对制剂中醋北柴胡、白芍、牡丹皮、炒栀子、丹参、甘草进行定性鉴别,在日光或紫外灯(254 nm或365 nm)下观察与对照品或对照药材相同位置是否有相同颜色的斑点;采用高效液相色谱法(HPLC)对方中所含芍药苷进行定量测定.色谱柱为Eclipse XDB-C18柱(250 mm×4.6 mm,5 μm);流动相为乙腈-0.1%磷酸溶液(体积比12∶88),柱温为25℃,流速为1.0 mL/min,检测波长为230 nm.结果 TLC鉴别中,各供试药材斑点显色清晰,与对照药材相同位置显相同颜色的斑点,且阴性对照无干扰.芍药苷进样量在0.131 2~1.180 8 μg范围内与峰面积之间线性关系良好,其回归方程为y=1×106x-18 220,R2=0.999 5.芍药苷定量限为0.087 μg、检测限为0.024 μg,稳定性、重复性试验结果的RSD均小于1.5%(n=6).平均加样回收率为95.95%,RSD为1.64%.3批样品中芍药苷平均转移率为56.60%.本品每袋含白芍、牡丹皮以芍药苷(C23H28O11)计,不得少于18.34 mg.结论 所建立的醋北柴胡、白芍、牡丹皮、炒栀子、丹参、甘草薄层色谱定性鉴别法与芍药苷HPLC含量测定方法操作简单、结果准确、专属性良好,可用于柴芍解郁颗粒的质量控制.
Objective To establish a quality standard for Chaishao Jieyu granules.Methods Thin-layer chromatography(TLC)was used for the qualitative identification of Bupleuri Radix Praeparata cum Aceto,Paeoniae Radix Alba,Moutan Cortex,Gardeniae Fructus Praeparatus,Salviae Miltiorrhizae Radix et Rhizoma,and Glycyrrhizae Radix et Rhizoma in the granules.Characteristic spots were observed at the positions corresponding to those of reference substances or reference herbs under daylight or ultraviolet light(254 nm or 365 nm).High-performance liquid chromatography(HPLC)was applied for the quantitative determination of paeoniflorin.The separa-tion was performed on an Eclipse XDB-C18 column(250 mm×4.6 mm,5 μm)with acetonitrile-0.1%phosphoric acid solution(volume ratio 12∶88)as the mobile phase.The column temperature was set at 25℃,the flow rate was 1.0 mL/min,and the detection wave-length was 230 nm.Results In TLC,clear and specific spots of all medicinal materials were observed at positions corresponding to the control samples,and no interference was found in negative controls.Paeoniflorin showed a good linear relationship in the range of 0.131 2-1.180 8 μg,with the regression equation of y=1×106x-18 220(R²=0.999 5).The limits of detection and quantitation were 0.024 μg and 0.087 μg,respectively.The RSD values for stability and repeatability were both below 1.5%(n=6).The mean recovery rate was 95.95%(RSD=1.64%),and the average transfer rate of paeoniflorin in three batches was 56.60%.Therefore,the content of paeoniflorin in each sachet showed no less than 18.34 mg(C₂₃H₂₈O₁₁),calculated based on Paeoniae Radix Alba and Moutan Cortex.Conclusion The established TLC methods and HPLC method are simple,accurate and specific,and can be effectively applied for the quality control of Chaishao Jieyu granules.
