BHMT在糖尿病性白内障晶状体中的表达及功能研究OA

目的 探讨甜菜碱高半胱氨酸甲基转移酶(BHMT)在糖尿病性白内障(DC)晶状体中的表达及其对高糖环境下晶状体上皮细胞同型半胱氨酸(Hcy)代谢、凋亡和氧化损伤的影响.方法 检测角膜移植供体捐献者(CL组)、年龄相关性白内障患者(ARC组)和DC患者(DC组)晶状体皮质及核部BHMT mRNA和蛋白表达以及Hcy水平(每组10枚晶状体).将人晶状体上皮细胞HLEC-B3分为对照组、高糖孵育组(HG组,30 mmol/L葡萄糖)、BHMT过表达组(BHMT-OE组)、BHMT过表达+高糖孵育组(BHMT-OE+HG组).各组细胞均培养24 h.用MTT法和EdU法检测HLEC-B3细胞增殖,TUNEL法检测HLEC-B3细胞凋亡,按试剂盒说明测定HLEC-B3细胞中Hcy和活性氧(ROS)水平及还原型谷胱甘肽(GSH)和氧化型谷胱甘肽(GSSG)活性.RT-qPCR检测BHMT和αA-晶状体蛋白(CRYAA)mRNA水平,Western blot检测BHMT、内质网应激(ERS)相关蛋白、B细胞淋巴瘤-2(Bcl-2)、CRYAA蛋白水平.结果 与CL组比较,ARC组和DC组晶状体皮质和核中的BHMT mRNA和蛋白水平降低,Hcy水平升高(均P<0.05).与ARC组比较,DC组晶状体皮质中的BHMT mRNA和蛋白水平降低,Hcy水平升高(均P<0.05);晶状体核中的BHMT mRNA和蛋白水平升高(P<0.05),Hcy水平降低(P<0.05).与对照组比较,HG组BHMT和CRYAA的mRNA和蛋白水平、相对细胞活力、EdU阳性率、Bcl-2蛋白水平和GSH含量降低(P<0.05),TUNEL阳性率、ROS相对荧光强度、Hcy和GSSG含量以及ERS相关蛋白水平升高(P<0.05).与HG组比较,BHMT-OE+HG组BHMT和CRYAA的mRNA和蛋白水平、相对细胞活力、EdU阳性率、Bcl-2蛋白水平和GSH含量升高(P<0.05),TUNEL阳性率、ROS相对荧光强度、Hcy和GSSG含量以及ERS相关蛋白水平降低(P<0.05).结论 BHMT在DC患者晶状体中下调,上调BHMT通过调控Hcy代谢、缓解氧化应激和内质网应激、上调CRYAA表达促进晶状体上皮细胞增殖、抑制凋亡.

Objective To investigate the expression of betaine-homocysteine methyltransferase(BHMT)in the lens of diabetic cataract(DC)and its effect on homocysteine(Hcy)metabolism,apoptosis and oxidative damage in lens epithelial cells exposed to high glu-cose condition.Methods Lens samples were collected from corneal transplant donors(CL group),age-related cataract patients(ARC group),and diabetic cataract patients(DC group),with 10 specimens in each group.The mRNA and protein expression of BHMT,as well as Hcy levels,were detected in in the lens cortex and nucleus of these samples.Human lens epithelial cells HLEC-B3 were divided into four groups:control group,high glucose incubation group(HG group,30 mmol/L glucose),BHMT overexpression group(BHMT-OE group),and BHMT overexpression plus high glucose incubation group(BHMT-OE+HG group).Cells in each group were cultured for 24 hours.Cell proliferation of HLEC-B3 cells was accessed by MTT assay and EdU assay,and cell apoptosis by TUNEL assay.The levels of Hcy and reactive oxygen species(ROS),as well as the activities of reduced glutathione(GSH)and oxidized gluta-thione(GSSG),were measured using commercial kits following the manufacturers' instructions.The mRNA levels of BHMT and αA-crystallin(CRYAA)were detected by RT-qPCR.Western blot was applied to measure the protein levels of BHMT,endoplasmic reticu-lum stress(ERS)-related proteins,B-cell lymphoma-2(Bcl-2)and CRYAA.Results Compared with the CL group,the mRNA and protein levels of BHMT in the lens cortex and nucleus were decreased,while Hcy levels were increased in both the ARC group and the DC group(all P<0.05).Compared with the ARC group,the mRNA and protein levels of BHMT in the lens cortex were decreased and the Hcy level was increased in the DC group(all P<0.05);in contrast,in the lens nucleus of the DC group,the mRNA and protein levels of BHMT were increased(P<0.05),while Hcy levels were decreased(P<0.05).Compared with the control group,the mRNA and protein levels of BHMT and CRYAA,the relative cell viability,EdU positive rate,Bcl-2 protein level and GSH content in the HG group were decreased(P<0.05);conversely the TUNEL positive rate,relative fluorescence intensity of ROS,contents of Hcy and GSSG,and levels of ERS-related proteins were increased(P<0.05).Compared with the HG group,the mRNA and protein levels of BHMT and CRYAA,the relative cell viability,EdU positive rate,Bcl-2 protein level and GSH content in the BHMT-OE+HG group were increased(P<0.05);however the TUNEL positive rate,relative fluorescence intensity of ROS,contents of Hcy and GSSG,and levels of ERS-related proteins were decreased(P<0.05).Conclusion BHMT is down-regulated in the lenses of DC patients.U-pregula-tion of BHMT promotes lens epithelial cells proliferation and inhibits apoptosis by modulating Hcy metabolism,alleviating oxidative and endoplasmic reticulum stress(ERS),and upregulating CRYAA expression.
