亚硫酸钠对与脂肪细胞共培养的肝细胞脂肪代谢的影响OA
Effect of sodium sulfite on hepatic lipid metabolism in a hepatocyte-adipocyte co-culture system
目的 探讨亚硫酸钠(Na2SO3)是否通过干扰脂肪细胞脂联素分泌进而影响肝细胞的脂肪代谢.方法 将3T3-L1前脂肪细胞诱导分化为成熟脂肪细胞.用完全培养基(阴性对照)、不同浓度Na2SO3(0.02,0.1,0.5,2.5 mmol/L)和1 mmol/L油酸(OA,阳性对照)处理脂肪细胞、单独培养HepG2细胞和共培养HepG2细胞.采用酶联免疫吸附法(ELISA)测定脂肪细胞培养上清中游离脂肪酸(FFA)和脂联素含量;采用甘油-3-磷酸氧化酶-过氧化物酶偶联法(GPO-POD)测定甘油三酯(TG)含量;采用免疫印迹法(Western blot)检测TG代谢相关蛋白固醇调节元件结合蛋白-1c(SREBP-1c)、乙酰辅酶A羧化酶(ACC)、脂肪酸合成酶(FASN)、腺苷酸活化蛋白激酶(AMPKα)、肉碱酯酰转移酶1(CPT-1)的相对表达水平.结果 与阴性对照相比,Na2SO3各处理组脂肪细胞培养上清中FFA含量明显减少(P<0.001),脂联素的含量明显减少(P<0.01).2.5 mmol/L Na2SO3可引起共培养HepG2细胞内TG含量增加(P<0.001).Western blot结果显示,在单独培养组,与阴性对照相比,各浓度Na2SO3处理后,SREBP-1c、p-ACC、AMPKα、CPT-1无明显变化(P>0.05);2.5 mmol/L Na2SO3处理后,ACC和FASN蛋白表达增高(P<0.05).在共培养组中,与阴性对照相比,各浓度Na2SO3 处理组ACC和AMPKα的蛋白表达均增高(P<0.05).结论 较低剂量Na2SO3暴露不会引起肝细胞内TG的增加.较高剂量的Na2SO3(2.5 mmol/L)可能通过对脂肪细胞脂联素分泌的抑制作用引起肝细胞中TG明显升高.
Objective To investigate whether sodium sulfite(Na2SO3)affects lipid metabolism in hepatocytes by interfering with adiponectin secretion from adipocytes.Methods 3T3-L1 preadipocytes were induced to differentiate into mature adipocytes.Adipo-cytes,HepG2 cells,and their co-cultured cells were treated with complete medium(negative control),different concentrations of Na2SO3(0.02,0.1,0.5,2.5 mmol/L),and 1 mmol/L oleic acid(OA,positive control),respectively.The levels of free fatty acid(FFA)and adiponectin in adipocyte culture supernatants were measured using enzyme-linked immunosorbent assay(ELISA),and triglyceride(TG)content was measured by the glycerol-3-phosphate oxidase-peroxidase(GPO-POD)method.Western blot was used to detect the relative expressions of TG metabolism-related proteins:sterol regulatory element-binding protein-1c(SREBP-1c),acetyl-CoA carboxylase(ACC),fatty acid synthase(FASN),adenosine monophosphate-activated protein kinase(AMPKα),and carnitine palmitoyltransferase 1(CPT-1).Results Compared with negative control group,FFA levels in the adipocyte culture supernatant were significantly reduced in all Na2SO3 treatment groups(P<0.001),and adiponectin levels were significantly decreased(P<0.01).The 2.5 mmol/L Na2SO3 caused an increase in TG content in co-cultured HepG2 cells(P<0.001).In HepG2 cells cultured alone,com-pared with the negative control,all concentrations of Na2SO3 did not significantly alter the expressions of SREBP-1c,p-ACC,AMPKα,and CPT-1(P>0.05),and 2.5 mmol/L Na2SO3 increased ACC and FASN protein expressions(P<0.05).In the co-cultured HepG2 cells,compared with the negative control,all concentrations of Na2SO3 increased protein expressions of ACC and AMPKα(P<0.05).Conclusion Exposure to low doses of Na2SO3 does not increase TG in hepatocytes.High doses of Na2SO3(2.5 mmol/L)may significantly elevate TG level in hepatocytes by inhibiting adiponectin secretion from adipocytes.
