[目的]克隆马铃薯甲虫触角中的气味受体基因(odorant receptor)OR30,明确 LdecOR30 在马铃薯甲虫雌雄成虫不同组织中的表达特性,及对寄主挥发物及虫体挥发物中主要醇类的电生理反应,解析 LdecOR30 的功能.[方法]在马铃薯甲虫全长转录组加二代转录组重测序的基础上,通过 PCR 技术克隆得到 LdecOR30 基因 CDS 序列;使用 qPCR 研究其在马铃薯甲虫头(无触角)、触角、足、翅、腹和胸不同组织中的表达特性;通过黑腹果蝇空神经元系统结合单感器记录研究 Lde-cOR30 对 16 种醇类化合物的电生理反应;使用 Y 型嗅觉仪测试特异反应化合物对马铃薯甲虫的行为影响.[结果]本研究克隆得到马铃薯甲虫 LdecOR30 cDNA 全长序列.序列分析显示,该气味受体含有 6 个跨膜结构域.qPCR 结果表明,其在雌雄成虫触角中的表达量显著高于其他组织.果蝇空神经元系统单感器记录结果显示,其对 5 种醇类产生的脉冲数量每秒增加超过 20 个.Y 型嗅觉仪选择试验显示,顺-3-己烯醇对马铃薯甲虫雌虫有显著引诱作用,引诱率为 80%,1-辛烯-3-醇对马铃薯甲虫雌雄成虫有显著的趋避作用,趋避率为 70%.[结论]马铃薯甲虫气味受体 LdecOR30 在雌雄成虫触角中均高表达,对顺-3-己烯醇等 5 种醇类有反应,推测其通过识别顺-3-己烯醇等醇类在马铃薯甲虫识别和定位寄主植物过程中发挥作用.

[Aim]This study aims to examine the function of LdecOR30 in the Colorado potato beetle,Leptinotarsa decemlineata.To do so,the odorant receptor gene OR30 from the antennae was cloned to determine the expression levels of LdecOR30 in different tis-sue samples of male and female adults,and investigate its electrophysiological responses to the main alcohols in various odor mole-cules.[Method]On the basis of the full-length transcriptome and next-generation transcriptome re-sequencing data of L.decemlin-eata,the coding sequence(CDS)of LdecOR30 was cloned by PCR.Quantitative real-time PCR(qPCR)was employed to analyze the expression profiles of LdecOR30 in the antennae,head(without antennae),thorax,abdomen,legs,and wings.The electrophys-iological responses of LdecOR30 to 16 alcohols were examined via the Drosophila empty neuron system combined with single sensil-lum recording.A Y-tube olfactometer was employed to test the effects of specifically responsive compounds on the behavior of L.de-cemlineata.[Result]The full-length cDNA sequence of LdecOR30 was successfully cloned.Sequence analysis indicated that this o-dorant receptor contained six transmembrane domains.qPCR results showed significantly higher expression of LdecOR30 in the anten-nae of both male and female adults than in other tissue samples.Single sensillum recordings in the Drosophila empty neuron system indicated that LdecOR30 responded to five alcohols,with an increase of over 20 spikes per second.Y-tube olfactometer assays dem-onstrated that cis-3-hexen-1-ol had a significant attractive effect on female beetles,with an attraction rate of 80%,while 1-octen-3-ol exhibited significant repellency of 70%to both male and female adults.[Conclusion]The odorant receptor gene LdecOR30 is highly expressed in the antennae of L.decemlineata adults,regardless of sex,and responds to five different alcohols,including cis-3-hexen-1-ol.These findings suggest that LdecOR30 plays an important role in the recognition and localization of host plants by L.de-cemlineata.
