两种玛草蛉的线粒体基因组全序列分析及草蛉科系统发育OA

为明确玛草蛉属Mallada 全线粒体基因组结构及其系统地位,基于二代测序技术对黄斑玛草蛉Mallada flavimaculus和绿玛草蛉Mallada viridianus的全线粒体基因组序列测序注释,并比较分析草蛉科线粒体基因组特征及系统发育关系.结果表明:黄斑玛草蛉和绿玛草蛉线粒体基因组序列总长分别为16 204 bp和15 810 bp,AT含量分别为80.14%和79.16%;大部分蛋白质编码基因的起始密码子为ATN,终止密码子以TAA为主,亦存在不完全终止形式(TA-tRNA或T-tRNA);ATP8核苷酸进化速率最高,COX1的核苷酸进化速率最低;除tRNASer(AGN)外,其余的tRNAs均可形成典型的三叶草结构.在草蛉科中,网蛉亚科与幻草蛉亚科+草蛉亚科构成姐妹群;在草蛉亚科中,Nothancylini是其余类群的姐妹群,亮草蛉族与尖草蛉族互为姐妹群,绢草蛉族是并系群,尼草蛉属Nineta Navás是草蛉族的姐妹群;玛草蛉属是通草蛉属的姐妹群.结果可为草蛉科分子系统学研究提供基础数据,更有助于阐述草蛉科线粒体基因组分化规律.

To clarify the complete mitochondrial genome(mitogenome)and systematic status of Mallada Navás,1925,the complete mitogenome of Mallada flavimaculus and Mallada viridianus were newly sequenced and annotated by Illumina second-generation sequencing technology.Furthermore,the characteristics of mitogenomes and the phylogenetic relationships within the family Chrysopidae were comparatively analyzed.The result showed that the complete mitogenome of M.flavimaculus and M.viridianus were 16 204 bp and 15 810 bp long,respectively.The AT content of the mitogenome of M.flavimaculus and M.viridianus were 80.14%and 79.16%.ATN was the mostly used start codon among the PCGs of mitogenome in Chrysopidae.Most PCGs used TAA as stop codons,while some PCGs terminated with TA-tRNA or T-tRNA.COX1 was the most conserved gene in the mitogenome of Chrysopidae,while ATP8 was a rapidly evolved gene.Almost all tRNAs could be folded into clover leaf structures,with the exception of tRNASer(AGN).Within Chrysopidae,Apochrysinae were recovered as the sister group to Chrysopinae+Nothochrysinae.Within Chrysopinae,Nothancylini were recovered as the sister group to(Leucochrysini+Belonopterygini)+(Ankylopterygini+(Nineta+Chrysopini)).Mallada were recovered as the sister group to Chrysoperla.These findings not only contribute foundational data for molecular systematic studies in Chrysopidae but also aid in elucidating the differentiation patterns of Chrysopidae mitogenomes.
