基于转录组学探讨天麻乙酸乙酯提取物治疗肌少症的作用机制OA
Transcriptomics-based Analysis of the Mechanism of Action of the EtOAc Extract from Gastrodia elata in the Treatment of Sarcopenia
目的 本研究基于转录组学,采用秀丽隐杆线虫复制地塞米松诱导的肌少症模型,探讨天麻乙酸乙酯提取物(EtOAc extract from Gastrodia elata,EEGE)对肌少症的防治作用.方法 将L4期的N2线虫置于相应实验板上,完成分组给药和模型的复制.采用游泳与爬行实验检测肌肉功能,鬼笔环肽染色观察肌肉状态.检测活性氧荧光染色和一氧化氮含量、抗氧化酶活性以及总抗氧化能力反映氧化应激水平.转录组学和PCR微阵列筛选研究EEGE治疗肌少症的可能机制.结果 与模型组相比,EEGE给药后游泳频次和爬行频次显著提高(P<0.01,P<0.001).EEGE给药后线虫体壁肌肉条纹清晰、排列规律.EEGE能够有效对抗氧化应激损伤,显著降低线虫体内活性氧和一氧化氮水平(P<0.001),并显著提高抗氧化酶活性和总抗氧化能力(P<0.05,P<0.001).转录组学分析表明,EEGE可能通过促进自噬、上调谷胱甘肽代谢相关通路以及调节细胞基质、角质层等生物过程,缓解肌少症.结论 EEGE能够显著改善线虫的肌肉功能和状态,对肌少症具有潜在的治疗作用.
Objective Based on transcriptomics,Caenorhabditis elegans was used to replicate a dexamethasone-induced sarcopenia model to explore the preventive and therapeutic effects of EtOAc extract from Gastrodia elata(EEGE)on sarcopenia.Methods L4-stage N2 worms were placed on corresponding experimental plates to complete drug administration and model replication.Muscle function was detected by swimming and crawling tests,and muscle state was observed by phalloidin staining.Reactive oxygen species(ROS)fluorescence staining,nitric oxide(NO)content,antioxidant enzyme activity,and total antioxidant capacity were measured to reflect the level of oxidative stress.Transcriptomics and PCR-based microarrays were used to screen for possible mechanisms of EEGE in treating sarcopenia.Results Compared with the model group,EEGE treatment significantly increased swimming and crawling frequencies(P<0.01,P<0.001).After EEGE treatment,the body wall muscle stripes of the worms were clear and regularly arranged.EEGE effectively combated oxidative stress damage,significantly reducing ROS and NO levels in the worms(P<0.001)while increasing antioxidant enzyme activity and total antioxidant capacity(P<0.05,P<0.001).Transcriptomics analysis suggested that EEGE may alleviate sarcopenia by promoting autophagy,upregulating glutathione metabolism-related pathways,and regulating biological processes such as the cell matrix and cuticle.Conclusion The results indicate that EEGE significantly improves muscle function and state in worms,and has potential therapeutic effects on sarcopenia.
