目的 利用质谱成像(Massspectrometry imaging,MSI)技术原位可视化与代谢组学技术的数据降维特点,揭示酸枣仁汤(Suanzaoren Decoction,SZRD)调控睡眠剥夺大鼠脑微区空间代谢网络而发挥抗失眠伴焦虑样的作用机制.方法 制备改良多平台水环境睡眠剥夺大鼠模型,通过高架十字迷宫实验以及血清皮质酮水平评价及SZRD的药效作用;利用质谱成像技术,观察睡眠剥夺大鼠脑中小分子代谢物的原位表达以及变化趋势,探究SZRD的作用特点.结果 正常大鼠改良多平台法剥夺21天后,出现焦虑样行为.与模型组比,SZRD显著增加了大鼠进入开臂的时间比例以及次数(P<0.05),降低了血清皮质酮含量(P<0.01),显著改善了睡眠剥夺大鼠的失眠和焦虑样行为.MSI结果显示,慢性睡眠剥夺显著影响了大鼠海马、下丘脑、纹状体和皮层中神经递质及其代谢物的含量;SZRD显著回调了脑内34 种差异代谢物含量,主要包括氨基酸类及其代谢产物等.通路分析提示SZRD可能通过调控组氨酸代谢,丙氨酸、天冬氨酸与谷氨酸代谢,以及色氨酸代谢、酪氨酸代谢、精氨酸代谢通路而发挥作用.结论 SZRD通过脑中调控色氨酸、酪氨酸等小分子氨基酸的代谢通路以及脑中神经递质的含量以及空间位置分布发挥抗失眠伴焦虑的作用.研究结果为SZRD精准用药,多靶点改善睡眠障碍的作用特点提供了科学依据.

Objective To elucidate the mechanism by which Suanzaoren Decoction(SZRD)exerts its anti-insomnia and anxiolytic-like effects by in situ visualization of spatial metabolic networks in specific brain microregions of sleep-deprived rats,leveraging the data dimensionality reduction features of metabolomics and mass spectrometry imaging(MSI)technology.Methods A modified multiple-platform water environment rat model of sleep deprivation was established.The therapeutic efficacy of SZRD was evaluated using the elevated plus maze test and serum corticosterone levels.MSI technology was employed to observe the in situ expression and trends of small-molecule metabolites in the brains of sleep-deprived rats and to investigate the intervention characteristics of SZRD.Results After 21 days of sleep deprivation using the modified multiple-platform method,normal rats exhibited anxiety-like behaviors.Compared to the model group,SZRD significantly increased the time ratio and number of entries into the open arms(P<0.05),reduced serum corticosterone levels(P<0.01),and markedly ameliorated insomnia and anxiety-like behaviors in sleep-deprived rats.MSI results showed that chronic sleep deprivation significantly altered the levels of neurotransmitters and their metabolites in the hippocampus,hypothalamus,striatum,and cortex of rats.SZRD significantly modulated the levels of 34 differential metabolites in the brain,primarily including amino acids and their metabolites.Pathway analysis suggested that SZRD may exert its effects by regulating histidine metabolism,alanine,aspartate and glutamate metabolism,tryptophan metabolism,tyrosine metabolism,and arginine metabolism pathways.Conclusion SZRD exerts anti-insomnia and anxiolytic effects by regulating small-molecule amino acid metabolic pathways(such as tryptophan and tyrosine metabolism)and modulating the content and spatial distribution of neurotransmitters in the brain.These findings provide a scientific basis for the precise application and multi-target characteristics of SZRD in improving sleep disorders.
