目的:建立砷(As)暴露6个月小鼠模型,揭示慢性低剂量As暴露诱导肾毒性的作用.方法:16只小鼠按体质量随机分为对照组(饮用蒸馏水)和As暴露组,每组8只.As暴露组饮用含5 mg/L As的水溶液,均连续6个月.HE染色评价肾脏结构变化,生物化学法检测小鼠血清尿素氮(BUN)、肌酐(CR)、T-超氧化物歧化酶(T-SOD)和丙二醛(MDA)含量,实时荧光定量PCR法检测核因子-κB(NF-κB)及促炎因子肿瘤坏死因子-α(TNF-α)mRNA表达水平.结果:As暴露组小鼠肾小球毛细血管内少量充血,肾小管结构基本完整,但近曲小管上皮细胞有水肿,肾间质有炎性细胞浸润.表明As暴露产生炎性反应,引起小鼠肾组织结构损伤.与对照组比较,As暴露组小鼠血清BUN、CR水平及MDA含量明显增加,T-SOD活性明显下降,NF-κB和TNF-α mRNA 表达明显升高,差异均有统计学意义(P<0.01).结论:长期As暴露诱导小鼠肾脏产生氧化应激和炎性反应,NF-κB和TNF-α参与了肾脏的炎性反应.

Objective:To establish a mouse model exposed to arsenic for 6 months and elucidate the nephrotoxic ef-fects induced by chronic low-dose arsenic exposure.Methods:Sixteen mice were randomly divided into a control group(drinking distilled water)and an arsenic exposure group.The arsenic exposure group received water containing 5 mg/L arse-nic for 6 months.Renal structural changes were evaluated by HE staining.Blood urea nitrogen(BUN),creatinine(CR),total superoxide dismutase(T-SOD)activity,and malondialdehyde(MDA)levels in serum were measured by biochemical meth-ods.The mRNA expression levels of nuclear factor-κB(NF-κB)and the pro-inflammatory cytokine tumor necrosis factor-α(TNF-α)were detected by quantitative real-time PCR.Results:In the arsenic exposure group,the glomerular capillaries showed slight congestion,and the renal tubular structure was largely intact.However,edema was observed in the epithelial cells of the proximal convoluted tubules,and inflammatory cell infiltration was present in the renal interstitium.This indicates that arsenic exposure induced an inflammatory response and caused structural damage to the mouse kidneys.Compared with the control group,the arsenic exposure group showed significantly increased serum levels of BUN,CR,and MDA,signifi-cantly decreased T-SOD activity,and significantly elevated mRNA expression of NF-κB and the pro-inflammatory factor TNF-α.All differences were statistically significant(P<0.01).Conclusion:Long-term arsenic exposure induces oxidative stress and an inflammatory response in mouse kidneys,with NF-κB and TNF-α involved in the renal inflammatory response.
