寻常型银屑病患者血清miR-204-5p,CCN1表达水平变化及其与免疫炎症因子的关系OA
Changes in Serum miR-204-5p and CCN1 Expression in Patients with Psoriasis Vulgaris and Their Relationship with Immune Inflammatory Factors Response
目的 探究寻常型银屑病(PV)患者血清微小RNA-204-5p(miR-204-5p)和富含半胱氨酸的蛋白61(CCN1)表达变化及其与免疫炎症反应的关系.方法 选取在2022年8月至2024年8月在我院就诊且符合研究条件的80例PV患者为研究对象,并依据PV患者的患病情况将其分为轻度组44例和重度组36例,另选取同期来我院体检的60例健康者作为对照组.使用qRT-PCR法测量miR-204-5p在血清中的表达,使用ELISA实验检测血清中CCN1的表达;使用Pearson相关性分析血清中miR-204-5p和CCN1表达与免疫炎症有关指标的关系;使用Logistic回归分析免疫炎症相关因子对PV发生的影响.结果 轻度PV组和重度PV组血清中miR-204-5p表达显著低于对照组,且重度PV组低于轻度PV组(P＜0.05);轻度PV组和重度PV组血清中CCN1表达显著高于对照组,且重度PV组高于轻度PV组(P＜0.05).轻度PV组和重度PV组患者的ESR、中性粒细胞计数、Th1/Th2、IL-2、INF-γ、hs-CRP水平显著高于对照组,且重度PV组高于轻度PV组(P＜0.05);轻度PV组和重度PV组IL-4和IL-10水平显著低于对照组,且重度PV组低于轻度PV组(P＜0.05);重度PV组患者PASI、DLQI、BSA显著高于轻度PV组(P＜0.05).血清miR-204-5 p表达水平与ESR、中性粒细胞计数、Th1/Th2、IL-2、INF-γ、hs-CRP水平呈负相关,与IL-4和IL-10水平呈正相关(P＜0.05);血清中CCN1表达与ESR、中性粒细胞计数、Th1/Th2、IL-2、INF-γ、hs-CRP水平呈正相关,与IL-4和IL-10水平呈负相关(P＜0.05).Th1/Th2、IL-2、INF-γ、IL-4、IL-10、miR-204-5p和CCN1 是 PV 发生的影响因素(P＜0.05).结论 PV 患者血清中 miR-204-5 p 的表达降低,而CCN1表达升高,二者表达水平均与PV患病程度以及与PV患者的炎症反应密切相关.
Objective To investigate expression changes of serum microRNA-204-5p(miR-204-5p)and cysteine-rich protein 61(CCN1)in patients with psoriasis vulgaris(PV),and their relationship with immune inflammatory response.Methods 80 PV patients who visited our hospital between August 2022 and August 2024 and met research criteria were selected as study subjects.They were assigned into a mild group of 44 patients and a severe group of 36 patients based on their disease status.Addi-tionally,60 healthy individuals who underwent physical examinations at our hospital during the same period were selected as the control group.RT-PCR method was applied to measure the expression of miR-204-5p in serum.ELISA experiment was applied to detect the expression of CCN1 in serum.Pearson correlation was applied to analyze the relationship between miR-204-5p and CCN1 expression in serum and immune inflammation related indicators.Logistic regression was applied to analyze the impact of immune inflammation related factors on the occurrence of PV.Results The miR-204-5p in serum mild PV group and severe PV group was significantly lower than that of control group,and severe PV group was lower than mild PV group(P＜0.05).The CCN1 in serum mild PV group and severe PV group was significantly higher than that of control group,and severe PV group was higher than mild PV group(P＜0.05).The ESR,neutrophil count,Th1/Th2,IL-2,INF-γ,and hs-CRP levels in mild PV group and severe PV group were significantly higher than those in control group,and severe PV group was higher than mild PV group(P＜0.05).IL-4 and IL-10 in mild PV group and severe PV group were significantly lower than those in control group,and severe PV group was low-er than mild PV group(P＜0.05).The PASI,DLQI and BSA score in severe PV group were significantly higher than those in mild PV group(P＜0.05).Serum miR-204-5p was negatively correlated with ESR,neutrophil count,Th1/Th2,IL-2,INF-γ,hs-CRP,and positively correlated with IL-4 and IL-10(P＜0.05).Serum CCN1 was positively correlated with ESR,neutrophil count,Th1/Th2,IL-2,INF-γ,hs-CRP,and negatively correlated with IL-4 and IL-10(P＜0.05).Th1/Th2,IL-2,INF-γ,IL-4,IL-10,miR-204-5p,and CCN1 were risk factors of PV occurrence(P＜0.05).Conclusion The miR-204-5p in PV patients is decreased,while CCN1 is increased.The expression levels of both are closely related to the severity of PV and inflammatory response of PV patients.
