A Study on Occurrence Regularity of Mycelium Fruiting Bodies and Hyphal Growth Medium of Cortinarius sinensis

为保护宁夏贺兰山中华丝膜菌资源并推动其人工驯化,采用野外定位观测与室内试验观测两种方式展开探究.野外观测结果表明,该菌发生于贺兰山海拔2 100 m以上的半阴半阳坡山脊上的青海云杉纯林(偶混生山杨等),林间郁闭度为0.58~0.66、pH值为7.5~8.0,土壤为灰褐土或高山草甸土,地表枯落物厚度为1.4~2.0 cm,0~10 cm土层菌丝密集;日均气温在14~25℃、地温在8~18℃、年降水量>500 mm时易发生,8月下旬至9月初为发生高峰,透雨后连续2~3 d高温是大量发生的关键条件之一,同时测定了其生长依赖的土壤腐殖质碳氮营养构成.基于土壤腐殖质养分的测定结果,以马铃薯浸出液(代碳源)、蛋白胨(代氮源)及MgSO₄、KH2PO4设计正交试验,同时用人工气候室模拟野外生境开展室内培养试验,结果表明:以马铃薯浸出液150 g/L、蛋白胨3 g/L、MgSO42 g/L、KH2PO42 g/L为配方的的培养基接种菌褶可促使菌丝生长,为该菌保护与驯化提供支撑.

To protect the Cortinarius sinensis in the Helan mountains of Ningxia and promote its artificial domestication,two methods of field observation and laboratory experiment observation were adopted for the investigation.The field observation results showed that this fungus occurred in the pure Picea crassifolia forest(occasionally mixed with Populus davidian,etc.)on the semi-shaded and semi-sunny slopes above an altitude of 2,100 meters in the Helan mountains,with a canopy density of 0.58 to 0.66,a pH value of 7.5 to 8.0,soil being gray-brown soil or alpine meadow soil,surface litter thickness of 1.4 to 2.0 cm,and dense mycelium in the 0 to 10 cm soil layer.It was prone to occur when the daily average temperature was 14 to 25℃,the ground temperature was 8 to 18℃,and the annual precipitation was more than 500 mm.The peak occurrence period was from late August to early September.The continuous 2 to 3 days of high temperature after a rain shower was one of the key conditions for a large number of occurrences.Meanwhile,the composition of soil humus carbon and nitrogen nutrients that the fungus relied on was determined.Based on the determination results of soil humus nutrients,orthogonal experiments were designed using potato extract solution(as a carbon source),peptone(as a nitrogen source),MgSO₄,and KH₂PO₄,and indoor cultivation experiments were carried out using an artificial climate chamber to simulate the wild habitat.The results showed that the culture medium with 150 g/L potato extract solution,3 g/L peptone,2 g/L MgSO₄,and 2 g/L KH₂PO₄ could promote the growth of mycelia when inoculated with the gill of this fungus,providing support for the protection and domestication of this fungus.
