苜蓿花叶病毒(AMV)LAMP检测方法的建立与检验OA
Establishment and Validation of a LAMP Detection Method for Alfalfa Mosaic Virus(AMV)
为建立快速、灵敏、可视化的苜蓿花叶病毒(Alfalfa mosaic virus,AMV)检测技术,基于AMV外壳蛋白(CP)基因高度保守序列设计特异性LAMP引物,通过引物筛选、反应条件优化,构建基于SYBR Green Ⅰ染料的可视化LAMP检测体系,并验证其特异性、灵敏度及田间应用效果.结果表明,优化后的最佳反应条件为65℃恒温反应60 min,内外引物浓度比8∶1,dNTPs浓度0.8 mmol/L,Mg2+浓度6.0 mmol/L.该体系对AMV产生特异性扩增,与苜蓿矮化病毒(ADV)、菜豆卷叶病毒(BLRV)无交叉反应,表现出良好的特异性.灵敏度检测显示,该方法的最低检测限为50 pg/μL,较常规RT-PCR灵敏度提高10倍.应用所建方法对20份田间疑似病样检测阳性检出率为85%,与RT-PCR验证结果一致.该检测方法操作简便、快速高效、成本较低,无需依赖复杂仪器设备,适用于基层实验室及田间现场快速检测,为AMV的早期预警、种苗检疫及病害防控提供了可靠的技术支撑.
To establish a rapid,sensitive,and visual detection method for Alfalfa mosaic virus(AMV),specific LAMP primers were designed based on highly conserved sequences of the AMV coat protein(CP)gene.Through primer screening and reaction condition optimization,a visual LAMP detection system based on SYBR Green Ⅰ dye was devel-oped,and its specificity,sensitivity,and field application efficacy were validated.The results indicated that the opti-mized reaction conditions were as follows:a 60-minute isothermal reaction at 65 ℃,an internal-to-external primer con-centration ratio of 8∶1,a dNTP concentration of 0.8 mmol/L,and a Mg2+concentration of 6.0 mmol/L.The system pro-duced specific amplification for AMV and showed no cross-reaction with Alfalfa Dwarfing Virus(ADV)or Bean Leaf Roll Virus(BLRV),demonstrating excellent specificity.Sensitivity testing revealed that the method's lower limit of de-tection was 50 pg/μL,representing a 10-fold increase in sensitivity compared to conventional RT-PCR.When applied to 20 field samples suspected of infection,the positive detection rate of the established method was 85%,showing high consistency with RT-PCR validation results.This detection method is simple to operate,rapid,efficient,and cost-effec-tive,requiring no complex instrumentation.It is suitable for rapid testing in grassroots laboratories and field settings,providing reliable technical support for early warning of AMV,seedling quarantine,and disease control.
