研究旨在明确新疆石河子地区规模化鸡场空肠弯曲杆菌的流行情况及其生物学特性,从当地4个规模化鸡场采集肛拭子样本198份,并通过形态学、生化鉴定、多重PCR方法进行分离株种属鉴定.采用K-B纸片扩散法开展药敏试验,PCR法检测耐药基因、毒力基因,结晶紫染色法评估生物被膜形成能力,开展小鼠致病性试验.结果显示:198份肛拭子样中分离出8株空肠弯曲杆菌,分离率为4.04%.3株分离株具有较强的生物被膜形成能力.8株分离株均为多重耐药,且对复方新诺明的耐药率为100.0%,而对头孢噻肟、恩诺沙星、阿奇霉素、红霉素、链霉素不耐药.耐药基因检测结果显示,L22的检出率最高(75.0%,6/8),aph(2'')-If的检出率最低(25.0%,2/8);毒力基因检测结果显示,wlaN、pldA的检出率最高(62.5%,5/8).分离株感染的小鼠1~3 d内全部死亡,心脏、肝脏、脾脏、肺脏、肾脏均出现明显的病理变化.研究表明,新疆石河子地区规模化鸡场空肠弯曲杆菌存在较高的耐药性及致病性,试验结果为空肠弯曲杆菌的防控提供了一定参考.

This study aimed to investigate the prevalence and biological characteristics of Campylobacter jejuni in large-scale chicken farms in the Shihezi Area of Xinjiang.A total of 198 anal swab samples were collected from four local large-scale farms.Isolates were identified through morphological and biochemical characterization,as well as multiplex PCR.Antimicrobial susceptibility testing was performed using the K-B disc diffusion method.Drug resistance genes and virulence genes were detected via PCR.Biofilm formation ability was assessed using the crystal violet staining method,and a mouse pathogenicity test was conducted.The results showed that eight strains of Campylobacter jejuni were isolated from the 198 anal swab samples,with an isolation rate of 4.04%.Three of the isolates exhibited strong biofilm-forming ability.All eight isolates were multidrug-resistant,showing 100.0%resistance to trimethoprim-sulfamethoxazole,while no resistance was observed to cefotaxime,enrofloxacin,azithromycin,erythromycin,or streptomycin.Regarding drug resistance genes,the highest detection rate was for L22(75.0%,6/8),while the lowest was for aph(2'')-If(25.0%,2/8).Virulence gene analysis revealed that wlaN and pldA had the highest detection rates(62.5%,5/8).All mice infected with the isolates died within 1 to 3 days,and significant pathological changes were observed in the heart,liver,spleen,lungs,and kidneys.The study indicates that Campylobacter jejuni in large-scale chicken farms in the Shihezi Area of Xinjiang exhibits high levels of drug resistance and pathogenicity.These findings provide a reference for the prevention and control of Campylobacter jejuni.
