试验旨在探究忍冬藤提取物(LJE)作为饲料添加剂对动物的抗炎作用.研究考察LJE对小鼠RAW 264.7单核细胞的毒性作用及炎症因子含量的影响,并通过构建由脂多糖(LPS)诱导的小鼠急性肺损伤(ALI)模型验证LJE对ALI的预防效果.结果显示,与对照组相比,LJE浓度高于4 g/L时对RAW 264.7细胞的毒性作用极显著增强(P<0.01).经LPS造模处理后,细胞培养液中炎症因子含量较对照组均极显著升高(P<0.01).与LPS组相比,4、8 g/L的LJE给药组RAW 264.7细胞上清液肿瘤坏死因子-α(TNF-α)含量极显著升高(P<0.01),白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)的含量极显著降低(P<0.01).给药后,小鼠肺泡灌洗液中白细胞、中性粒细胞及巨噬细胞的数量及TNF-α、IL-6、IL-1β的含量相比模型组均极显著降低(P<0.01);显微镜观察可见小鼠肺间质轻度水肿,肺泡壁轻度增宽;仍有少量白细胞浸润,但浸润程度显著减轻;肺泡壁无显著充血,结构维持完整,整体炎症反应程度较轻.研究表明,LJE具有显著的抗炎、抗氧化及保护肺组织的作用,能够有效缓解ALI症状.

The experiment aimed to investigate the anti-inflammatory effect of the extract of Lonicera japonica(LJE)as a feed additive on animals.The study examined the toxicity of LJE on RAW 264.7 mononuclear cells and its influence on the content of inflammatory factors,and verified the preventive effect of LJE on acute lung injury(ALI)by constructing a mouse model of ALI induced by lipopolysaccharide(LPS).The results showed that when the concentration of LJE was higher than 4 g/L,the toxicity of LJE on RAW 264.7 cells was extremely enhanced(P<0.01).After LPS induction,the content of inflammatory factors in the cell culture medium was extremely higher than that of the control group(P<0.01).Compared with the LPS group,the supernatant of RAW 264.7 cells in the 4 and 8 g/L LJE treatment groups showed extremely significant increases in tumor necrosis factor-α(TNF-α)content(P<0.01),while the contents of interleukin-6(IL-6)and interleukin-1β(IL-1β)decreased extremely(P<0.01).After administration,the number of white blood cells,neutrophils,and macrophages in the bronchoalveolar lavage fluid of mice and the contents of TNF-α,IL-6,and IL-1β were extremely lower than those of the model group(P<0.01).Microscopic observation revealed mild interstitial edema in the lung interstitium,mild widening of the alveolar wall.There were still a few white blood cells infiltrating,but the infiltration degree was significantly reduced;there was no significant congestion of the alveolar wall,and the structure was maintained intact,with a relatively mild inflammatory response overall.The study demonstrated that LJE has significant anti-inflammatory,antioxidant,and protective effects on lung tissue,and can effectively alleviate the symptoms of ALI.
