研究旨在建立微小隐孢子虫(Cryptosporidium parvum)的PCR检测方法,根据GenBank中下载的微小隐孢子虫的GP60基因序列,选择一段高度保守区设计出特异性引物,通过程序优化建立微小隐孢子虫PCR检测方法,并进行敏感性、特异性、重复性试验及临床样品检测.结果显示,所建立的PCR方法可扩增出921 bp的微小隐孢子虫特异性基因片段,与预期大小相符;与产毒素性大肠杆菌K99、产气荚膜梭菌、艾美尔球虫、牛轮状病毒、牛冠状病毒均无交叉反应;将阳性样本倍比稀释后进行PCR检测,最低检测浓度为0.243 mg/L;组内、组间试验结果显示,该方法具有良好的重复性和稳定性.使用该方法对从黑龙江地区采集的411份腹泻牛粪便、肛拭子样本进行检测,共检出141份微小隐孢子虫阳性样本,总阳性率为34.31%.研究表明,试验建立的方法具有良好的特异性、敏感性与重复性,能够较好地用于临床样本的检测.

This study aimed to establish a PCR method for the detection of Cryptosporidium parvum.Based on the GP60 gene sequence of Cryptosporidium parvum downloaded from GenBank,a pair of specific primers was designed targeting a highly conserved region.The PCR method was established through program optimization,and its sensitivity,specificity,repeatability,and clinical sample detection performance were evaluated.The results showed that the established PCR method could amplify a specific 921 bp fragment of Cryptosporidium parvum,consistent with the expected size.No cross-reactions were observed with enterotoxigenic Escherichia coli K99,Clostridium perfringens,Eimeria sp.,bovine rotavirus,and bovine coronavirus.After serial dilution of positive samples,the minimum detection concentration was 0.243 mg/L.Intra-assay and inter-assay results demonstrated good repeatability and stability.Using this method,411 fecal and anal swab samples from diarrheic cattle collected in Heilongjiang Province were tested,and 141 samples were positive for Cryptosporidium parvum,with an overall positive rate of 34.31%.The study indicates that the established method has good specificity,sensitivity,and repeatability,and is well suited for the detection of clinical samples.
