1种载药水凝胶的制备及抗菌作用研究OA
Preparation and antibacterial activity of a drug-loaded hydrogel
为应对动物细菌性皮肤伤口感染及姜黄素水溶性差、稳定性低的应用瓶颈,研究旨在开发一种新型载药纳米凝胶并评价其抗菌性能.研究通过离子交联与席夫碱反应相结合的原位交联法,以壳聚糖微球与负载姜黄素的相变微球为前体,按4∶6体积比成功制备了姜黄素纳米凝胶,通过扫描电镜(SEM)观察其结构,并对其抗菌效果进行了考察.结果显示,姜黄素纳米凝胶具有结构致密、孔径均匀(10~100 μm)的三维多孔结构,对金黄色葡萄球菌标准株ATCC 29213及其小菌落变异株(SCVA 102)均表现出优于游离姜黄素的抑菌活性和持久性,其最低抑菌浓度(MIC值)均为250、250 mg/L,且能够维持72 h的稳定抑菌效果.研究表明,试验制备的姜黄素纳米凝胶具有良好的缓释性能与显著的体外抗菌活性,为治疗动物皮肤伤口感染及控制耐药菌提供了具有潜力的局部给药新途径.
To address the challenges of bacterial skin wound infections in animals and the poor water solubility and low stability of curcumin,this study aimed to develop a new type of drug-loaded nanogel and evaluate its antibacterial properties.The study utilized the in-situ crosslinking method combining ionic crosslinking and Schiff base reaction,using chitosan microspheres and curcumin-loaded phase-change microspheres as precursors,and successfully prepared curcumin nanogel at a volume ratio of 4∶6.The structure was observed using scanning electron microscopy(SEM),and its antibacterial effect was investigated.The results showed that the curcumin nanogel had a dense structure with uniform pore diameters(approximately 10~100 μm)in a three-dimensional porous structure,and exhibited superior antibacterial activity and persistence compared to free curcumin against the standard strain of Staphylococcus aureus ATCC 29213 and its small colony variant(SCVA 102).The minimum inhibitory concentration(MIC value)was 250 and 250 mg/L,and it maintained a stable antibacterial effect for 72 hours.The study demonstrated that the prepared curcumin nanogel has good sustained-release properties and significant in vitro antibacterial activity,providing a potential new local administration route for treating animal skin wound infections and controlling drug-resistant bacteria.
