叶绿素是植物体内的核心色素,在驱动光合作用为植物提供能量和植物着色等方面起着关键作用.SlBBX20(Solanum lycopersicum B-box zinc finger transcription factor 20,番茄 B-box锌指转录因子20)是番茄SlBBXs 家族的重要成员,在类胡萝卜素合成、光形态建成、花青素积累及低温胁迫响应等方面发挥作用.为探究 SlBBX20 是否参与叶绿素代谢调控及对番茄叶绿素合成是否有影响,本研究对基因敲除技术获得的番茄SlBBX20基因敲除株系(Cri-BBX20)进行研究,发现其叶片颜色变浅,叶绿素含量显著降低;对野生型(A57)和敲除株系进行转录组测序,发现差异基因主要富集到氨基酸生物合成(Biosynthesis of amino acids)与碳代谢(Carbon metabolism)两个通路中;同时对 8 个与叶绿素合成相关的基因进行了启动子预测,发现其启动子均含有能与 SlBBX20 结合的顺式作用元件.研究推测 SlBBX20 可能通过调控叶绿素生物合成途径中相关基因的转录,进而影响番茄叶绿素的生物合成.

Chlorophyll is an essential core pigment in plants,which plays a vital role in driving photosynthesis to provide energy for plant growth and regulating plant coloration.SlBBX20(Solanum lycopersicum B-box zinc finger transcription factor 20)is a key member of the tomato SlBBXs gene family,and participates in carotenoid biosynthesis,photomorphogenesis,anthocyanin accumulation and low-temperature stress response.To clarify whether SlBBX20 regulates chlorophyll metabolism and influences chlorophyll synthesis in tomato,we investigated the SlBBX20 knockout line Cri-BBX20 generated by gene editing.The results showed that the knockout line exhibited pale leaf color and significantly decreased chlorophyll content.Transcriptome sequencing was performed using wild-type A57 and the knockout line.Functional enrichment analysis indicated that differentially expressed genes were mainly enriched in the biosynthesis of amino acids and carbon metabolism pathways.In addition,promoter cis-element prediction was conducted on eight chlorophyll synthesis-related genes,and the promoters of these genes all contained cis-acting elements that can bind to SlBBX20.Collectively,SlBBX20 may modulate the transcription of genes involved in the chlorophyll biosynthesis pathway,thereby regulating chlorophyll biosynthesis in tomato.
