拟穴青蟹钙调蛋白基因的分子特性及蜕皮作用分析OA
Molecular Characterizations of Calmodulin Gene and Analysis of Molting Process in Scylla paramamosain
克隆获得拟穴青蟹(Scylla paramamosain)钙调蛋白(calmodulin,CaM)基因的全长 cDNA(SpCaM),其长度为1980 bp,开放阅读框(open reading frame,ORF)为450 bp.ClustalW 序列比对和系统进化树结果显示,SpCaM 与甲壳动物 CaM 相似性最高,亲缘关系最近;实时荧光定量 PCR 结果显示,在仔蟹蜕皮周期中,SpCaM 在蜕皮间期 mRNA 表达量最高,且与蜕皮抑制激素基因(SpMIH)表达模式相似;RNA 干扰实验表明,仔蟹注射ds SpMIH48 h 后,SpMIH 与SpCaM 转录水平均显著降低.这些结果表明,SpCaM 参与拟穴青蟹蜕皮过程,且其表达可能受到 SpMIH 的调控.
In this study,the CaM gene was cloned from the mud crab Scylla paramamosain.The full-length cDNA of SpCaM was 1980 bp,and its 450 bp open reading frame(ORF)was isolated and characterized.Clust-alW and phylogenetic analysis showed that SpCaM was closely related to the CaM proteins from other crustace-ans.Quantitative real-time PCR revealed that during molt cycle of early juvenile S.paramamosain,the SpCaM mRNA levels reached the peak at the intermolt stage,and its expression pattern was similar to that of SpMIH.Further RNA interference experiment showed that injection of double stranded SpMIH(ds SpMIH)into the juven-ile crabs resulted in significant reduction of SpMIH and SpCaM transcripts at 48 h post injection.These findings indicated that SpCaM participates in regulating molting of S.paramamosain and was likely regulated by SpMIH.
