目的 本研究探讨肉豆蔻五味对心肌缺血大鼠心功能及脑损伤的影响.方法 选取32 只8 周龄雄性 SD 大鼠随机分为空白组、空白给药组、模型组和模型给药组,每组8 只.模型组及模型给药组采用冠状动脉左前降支结扎法制备心肌缺血模型,模型给药组于造模前一周给予肉豆蔻五味(0.54 g/kg)预处理.造模成功后,空白给药组及模型给药组给予相同剂量肉豆蔻五味进行干预,空白组及模型组给予等体积羧甲基纤维素钠溶液,连续干预 3 周.观察各组大鼠干预前后体质量变化、心电图情况;采用超声心动图检测心脏结构与功能;通过平衡木实验和水迷宫实验评估感觉运动功能及空间学习记忆能力;采用酶联免疫吸附法检测海马组织中白细胞介素-1β(interleukin-1β,IL-1β)、肿瘤坏死因子 α(tumor necrosis factor-α,TNF-α)、乙酰胆碱酯酶(acetylcholinesterase,AchE)及神经胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)的含量.结果 (1)体质量:与空白组比较,模型组大鼠术后第1 周出现"体质量假性升高",随后体质量持续下降(P<0.01),给药后体质量显著上升.(2)心电图:模型组大鼠 ST 段明显抬高,给药干预后基本恢复正常.(3)超声心动图:与空白组相比,模型组左心室前壁变薄,左心室舒张末期容积、左心室收缩末期容积显著增大(P<0.01),射血分数、左心室短轴缩短率显著降低(P<0.01);模型给药组上述指标均显著改善(P<0.01).(4)行为学:与空白组相比,模型组大鼠平衡木穿越时间和水迷宫逃避潜伏期显著延长(P<0.01);与模型组比较,模型给药组上述时间均显著缩短(P<0.01).(5)因子含量:与模型组相比,模型给药组海马组织中 IL-1β、TNF-α、AchE 及 GFAP含量均显著降低(P<0.05,P<0.01).结论 肉豆蔻五味能显著改善心肌缺血大鼠心脏功能,并可能通过抑制海马神经炎症、调节胆碱能系统及胶质细胞活化,减轻心肌缺血引起的认知功能与感觉运动功能障碍.

Objective This study investigated the effects of Nutmeg-5 on cardiac function and brain damage in rats with myocardial ischemia.Methods Thirty-two 8-week-old male SD rats were randomly divided into four groups(n=8 each):the control group,the control-treated group,the model group,and the model-treated group.Myocardial ischemia was induced in the model group Control-treated group via ligation of the left anterior descending coronary artery.The model drug group received pre-treatment with Nutmeg-5 extract(0.54 g/kg)one week prior to modeling.Following successful modeling,the model drug group and model drug group continued to receive the same dose of the drug,while the Normal Group and Model Group received an equal volume of carboxymethyl cellulose sodium solution,with continuous intervention for 3 weeks.Body Weight changes and electrocardiograms were monitored in all groups before and after intervention;Cardiac structure and function were assessed using cardiac echocardiography;sensorimotor function and spatial learning and memory abilities were evaluated via the balance beam test and water maze test;levels of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),acetylcholinesterase(AchE),and glial fibrillary acidic protein(GFAP)in hippocampal tissue were measured using the ELISA method.Results(1)Body Weight:Rats in the model group ex-hibited pseudo-weight gain in the first week post-surgery,followed by a continuous decline in body weight(compared with the control group,P<0.01);body weight showed an upward trend after drug administration.(2)Electrocardiogram:Rats in the model group exhibited significant ST-segment elevation,which largely returned to normal after drug intervention.(3)Echocardiography:Compared with the control group,the model group exhibited thinning of the left ventricular anterior wall,with significantly increased left ventricular end-diastolic volume and left ventricular end-systolic volume(P<0.01)and significantly decreased ejection fractions and fractional shortening(P<0.01);in the model group treated with the drug,all these parameters improved significantly(P<0.01).(4)Behavioral Tests:Compared with the control group,rats in the model group showed significantly prolonged balance beam crossing time and water maze escape latency(P<0.01);compared with the model group,the drug-treated model group exhibited significant reductions in these times(P<0.01).(5)Hippocampal Tissue Factors:Compared with the model group,the levels of IL-1β,TNF-α,AchE,and GFAP in the hippocampal tissue of the model-treated group were significantly reduced(P<0.05,P<0.01).Conclusion Nutmeg-5 significantly improves cardiac function in rats with myocardial ischemia and may alleviate sensory-motor and cognitive behavioral impairments caused by myocardial ischemia by inhibiting hippocampal neuroinflammation,modulating the cholinergic system,and regulating glial cell activation.
