基于电阻抗图谱法的月季白粉病检测技术研究OA

月季(Rosa chinensis)是世界上重要的鲜切花和园林应用植物,月季白粉病(Podosphaera pannosa)极大影响了月季的观赏和经济价值.为了降低月季白粉病的危害,加强病情早期检测和预警的能力,以2 a生月季高抗病品种"仙境"和易感病品种"红帽"为试材,进行月季白粉病菌接种处理,测定月季叶片的电阻抗图谱(Electrical impedance spectroscopy,EIS)和参数、病程相关蛋白酶活性、淀粉和可溶性糖含量等指标,分析EIS参数与生理指标的相关性,探究适宜月季白粉病的EIS参数,以便为建立快速检测月季白粉病提供新的技术方法.研究结果表明:(1)随着2个品种感病程度的加深,其电阻抗图谱弧度逐渐增大.其中易感病品种"红帽"在接种白粉病菌后3 d时与对照产生明显差异,高抗病"仙境"在接种白粉病菌后5 d时与对照产生明显差异.(2)未接种白粉病,高抗病品种"仙境"对照的几丁质酶、β—1,3—葡聚糖酶、多酚氧化酶活性高于易感病品种"红帽"对照酶活性,表明高抗性品种具有较高的病程相关蛋白酶活性.(3)接种白粉病菌后,高抗病品种"仙境"的几丁质酶、β—1,3—葡聚糖酶、多酚氧化酶活性均高于易感病品种"红帽",且2个品种的酶活性均高于对照.(4)接种白粉病菌后,2个品种的EIS参数均与生理参数相关,其中弛豫时间分布系数φ与生理参数均极显著相关,与几丁质酶活性和可溶性糖含量相关系数均达到r=0.688(P<0.01),与β-1,3—葡聚糖酶活性相关系数达到r=0.555(P<0.01),可以将其作为反映月季抗白粉病能力的最佳电阻抗参数.

Rosa chinensis is an important cut flower and garden plant worldwide.Podosphaera pannosa,the cause of rose powdery mildew,significantly affects the ornamental and economic value of roses.To reduce the damage caused by rose powdery mildew and enhance the early detection and warning capabilities of the disease,two-year-old rose varieties"Fairyland"(highly resistant)and"Red Hat"(susceptible)were used as test materials.They were inoculated with Podosphaera pannosa,and the electrical impedance spectroscopy(EIS)spectra and parameters,activities of pathogenesis-related proteases,and contents of starch and soluble sugar in rose leaves were measured.The correlations between EIS parameters and physiological indicators were analyzed to explore suitable EIS parameters for rose powdery mildew,providing a new technical method for rapid detection of rose powdery mildew.The results showed that:(1)As the disease severity increased in both varieties,the arc of the EIS spectra gradually increased.The susceptible variety"Red Hat"showed significant differences from the control 3 days after inoculation,while the highly resistant variety"Fairyland"showed significant differences from the control 5 days after inoculation.(2)Before inoculation,the activities of chitinase,β-1,3-glucanase,and polyphenol oxidase in the highly resistant variety"Fairyland"were higher than those in the susceptible variety"Red Hat",indicating that the highly resistant variety had higher activities of pathogenesis-related proteases.(3)After inoculation,the activities of chitinase,β-1,3-glucanase,and polyphenol oxidase in the highly resistant variety"Fairyland"were higher than those in the susceptible variety"Red Hat",and the activities of both varieties were higher than those in the control.(4)After inoculation with powdery mildew fungus,the EIS parameters of both varieties showed significantly correlated with physiological parameters.Ψ had was highly significantly correlated with physiological parameters,with correlation coefficients of r=0.688(P<0.01)for both chitinase activity and soluble sugar content,and r=0.555(P<0.01)for β-1,3-glucanase activity.Therefore,Ψ can be used as the best EIS parameter to reflect the resistance of rose to powdery mildew.
