吡非尼酮对三阴性乳腺癌4T1细胞增殖、迁移和侵袭影响的机制研究OA
Effects of pirfenidone on the proliferation,migration,and invasion of triple-negative breast cancer 4T1 cells
目的:探究吡非尼酮(pirfenidone,PFD)对三阴性乳腺癌4T1细胞增殖、迁移和侵袭能力影响的机制.方法:培养三阴性乳腺癌4T1细胞株,将不同浓度PFD(2、4、6、8 mmol/L)处理4T1细胞作为药物处理组,未加 PFD 的处理 4T1 细 胞 为 对 照 组.Western blot 检测芳胺 N-乙酰转移酶 1(arylamine N-acetyltransferase 1,NAT1)蛋白的表达;CCK-8实验和克隆形成实验检测PFD对4T1细胞增殖能力的影响;划痕实验和Transwell迁移实验检测PFD对4T1细胞迁移能力的影响;Transwell侵袭实验检测PFD对4T1细胞侵袭能力的影响;分子对接验证 PFD 与 NAT1之间的相互作用.结果:与对照组相比,PFD 在8 mmol/L浓度时明显抑制NAT1蛋白的表达(P<0.000 1);与对照组相比,经2、4、6、8 mmol/L PFD处理的4T1细胞增殖、迁移、侵袭能力逐渐减弱,具有浓度依赖性;其 中,PFD浓度为8 mmol/L时明显抑制了4T1细胞的增殖、迁移与侵袭能力(P<0.001);此外,分子对接显示,PFD与NAT1之间存在氢键及疏水相互作用.结论:8 mmol/LPFD通过抑制NAT1蛋白表达降低三阴性乳腺癌4T1细胞增殖、迁移和侵袭能力.
Objective:To explore the effects of of pirfenidone(PFD)on the proliferation,migration,and invasion abilities of triple-negative breast cancer 4T1 cells and its mechanisms.Methods:The triple-negative breast cancer 4T1 cell line was cultured.The 4T1 cells treated with different concentrations of PFD(2,4,6,and 8 mmol/L)were taken as the drug treatment group,and the 4T1 cells not treated with PFD were taken as the control group.The expression of arylamine N-acetyltransferase 1(NAT1)protein was detected by Western blot.The effect of PFD on 4T1 cells' proliferation ability was detected by CCK-8 assay and clone formation assay.The scratch assay and Transwell migration assay were used to detect the effect of PFD on 4T1 cells' migration ability.The Transwell invasion assay was used to detect the effect of PFD on 4T1 cells' invasion ability.Molecular docking was used to verify the interaction between PFD and NAT1.Results:Compared to the control group,PFD significantly inhibited the protein expression of NAT1 at the concentration of 8 mmol/L(P<0.000 1);Compared to the control group,the proliferation,migration,and invasion abilities of 4T1 cells treated with PFD at 2,4,6 and 8 mmol/L gradually weakened in a concentration-dependent manner.Among them,when the concentration of PFD was 8 mmol/L,it significantly inhibited the proliferation,migra-tion,and invasion abilities of 4T1 cells(P<0.001);Furthermore,molecular docking indicated that there were hydrogen bonds and hydrophobic interactions between PFD and NAT1.Conclusion:The concentration of PFD at 8 mmol/L reduces the prolifera-tion,migration,and invasion abilities of triple-negative breast cancer 4T1 cells by inhibiting the expression of NAT1 protein.
