[目的]观察中医经典方剂四君子汤对肝癌大鼠的治疗作用,并从肉瘤病毒癌基因同源物(Ras)/细胞外调节蛋白激酶(ERK)信号通路的角度分析其潜在机制.[方法]将50只SD大鼠随机分成5组,即正常组,模型组,四君子汤低、高剂量组和四君子汤高剂量+ML-099(Ras/ERK通路激活剂)组,每组10只.除正常组,其他各组大鼠采用腹腔注射二乙基亚硝胺(DEN)法构建肝癌模型.造模成功后,分组干预.干预后,计算各组大鼠肝指数.酶联免疫吸附分析(ELISA)检测血清肝功能指标[丙氨酸氨基转移酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)]及炎症因子[白细胞介素(IL)-1α、趋化因子2(CCL-2)和IL-6]水平.苏木精-伊红(HE)染色法观察肝脏组织的病理学变化.蛋白免疫印迹(Western Blot)法检测肝脏组织细胞凋亡相关蛋白剪切型半胱氨酸天冬氨酸蛋白酶 3(cleaved-Caspase 3)、B细胞淋巴瘤/白血病-2基因(Bcl-2)、Bcl-2相关X蛋白(Bax)和Ras/ERK信号通路相关蛋白Ras和磷酸化(p)-ERK/ERK的表达水平.[结果]与正常组比较,模型组大鼠的肝指数,ALT、AST、ALP、IL-1α、CCL-2、IL-6含量及cleaved-Caspase 3、Bcl-2、Ras和p-ERK/ERK的表达水平显著升高(P<0.05),而Bax的表达水平显著降低(P<0.05).与模型组比较,四君子汤低、高剂量组大鼠的肝指数,ALT、AST、ALP、IL-1α、CCL-2和IL-6含量及cleaved-Caspase 3、Bcl-2、Ras和p-ERK/ERK的表达水平显著降低(P<0.05),Bax的表达水平显著升高(P<0.05),呈剂量依赖性.与四君子汤高剂量组比较,四君子汤高剂量+ML-099组大鼠的肝指数,ALT、AST、ALP、IL-1α、CCL-2和IL-6含量及cleaved-Caspase 3、Bcl-2、Ras和p-ERK/ERK的表达水平显著升高(P<0.05),Bax的表达水平显著降低(P<0.05).[结论]四君子汤能够改善肝癌大鼠肝功能、减轻肝损伤,其作用机制可能与抑制Ras/ERK信号通路、缓解炎症反应并促进癌细胞凋亡有关.

Objective To observe the therapeutic effect of the classical traditional Chinese medicine formula Sijunzi Decoction on rats with hepatocellular carcinoma(HCC)and to analyze its potential mechanism from the perspective of the rat sarcoma viral oncogene homolog(Ras)/extracellular signal-regulated kinase(ERK)signaling pathway.Methods Fifty SD rats were randomly divided into 5 groups:normal group,model group,low-dose Sijunzi Decoction group,high-dose Sijunzi Decoction group,and high-dose Sijunzi Decoction+ML-099(Ras/ERK pathway activator)group,with 10 rats in each group.Except for the normal group,rats in the other groups received intraperitoneal injections of diethylnitrosamine(DEN)to establish the HCC model.After successful modeling,the groups received the respective interventions.Following the intervention,liver index was calculated in each group.Serum levels of liver function indicators[alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP)]and inflammatory factors[interleukin(IL)-1α,chemokine ligand 2(CCL-2),and IL-6]were detected by enzyme-linked immunosorbent assay(ELISA).Hematoxylin-eosin(HE)staining was used to observe histopathological changes in liver tissue.The expression levels of apoptosis-related proteins[cleaved cysteine aspartate protease 3(cleaved-Caspase 3),B-cell lymphoma/leukemia-2 gene(Bcl-2)]and Ras/ERK signaling pathway-related proteins[Ras and phosphorylated(p)-ERK/ERK]in liver tissue were detected by Western Blot.Results Compared with the normal group,the model group exhibited significantly increased liver index,serum levels of ALT,AST,ALP,IL-1α,CCL-2,and IL-6,as well as elevated expression levels of cleaved-Caspase 3,Bcl-2,Ras,and p-ERK/ERK(P<0.05),while the expression level of Bax was significantly decreased(P<0.05).Compared with the model group,both the low-dose and high-dose Sijunzi Decoction groups showed significantly reduced,liver index,serum levels of ALT,AST,ALP,IL-1α,CCL-2,and IL-6,and decreased expression levels of cleaved-Caspase 3,Bcl-2,Ras,and p-ERK/ERK(P<0.05),along with a significantly increased expression level of Bax(P<0.05),in a dose-dependent manner.Compared with the high-dose Sijunzi Decoction group,the high-dose Sijunzi Decoction+ML-099 group demonstrated significantly increased,liver index,serum levels of ALT,AST,ALP,IL-1α,CCL-2,and IL-6,and elevated expression levels of cleaved-Caspase 3,Bcl-2,Ras,and p-ERK/ERK(P<0.05),while the expression level of Bax was significantly decreased(P<0.05).Conclusion Sijunzi Decoction can improve liver function and alleviate liver injury in rats with HCC,and its mechanism of action may be associated with inhibiting the Ras/ERK signaling pathway,ameliorating inflammatory responses,and promoting cancer cell apoptosis.
