黄连解毒汤调节2型糖尿病大鼠粪便乳酸杆菌的实验研究OA
Investigation on the Effect of Huanglian Jiedu Decoction on Fecal Lactobacillus in Type 2 Diabetic Rats
目的 通过荧光定量 PCR 探讨黄连解毒汤对2 型糖尿病模型大鼠肠道乳酸杆菌数量的影响.方法 采用随机分组法将60 只SD 大鼠分为空白组(10 只)及造模组(50 只),适应性喂养7d 后,造模组采用高脂饲料诱导联合链脲佐菌素造模,以造模后空腹血糖≥16.7 mmol·L-1作为糖尿病模型成功依据.将成模大鼠按随机数字表法分为5 组(n=10):模型组,黄连解毒汤高(6.30 g·kg-1)、中(3.15 g·kg-1)、低(1.58 g·kg-1)剂量组,对照组(二甲双胍 200 mg·kg-1).造模后第 2 天,黄连解毒汤各剂量组及对照组给予相应药物连续灌胃21 d,空白组与模型组大鼠同步接受等量生理盐水灌胃;末次干预后禁食12 h(允许自由饮水),经尾静脉尖端穿刺采集血液检测空腹血糖,同步采集新鲜粪便进行 DNA 提取及荧光定量PCR 检测分析.结果 模型组大鼠肠道乳酸杆菌含量明显低于空白组,而通过黄连解毒汤及二甲双胍治疗后乳酸杆菌含量均有所回升,其中黄连解毒汤低剂量组和对照组乳酸杆菌含量较模型组显著性升高(P<0.05).结论 黄连解毒汤可增加2 型糖尿病模型大鼠肠道乳酸杆菌的含量,其中低剂量组的作用最为显著.
Objective To investigate the effect of Huanglian Jiedu Decoction on the quantity of Lactobacillus in type 2 diabetes mellitus(T2DM)model rats by fluorescence quantitative PRC(qPCR).Methods 60 SD rats were randomly divided into a blank group(10 rats)and a model group(50 rats).After 7 days of adaptive feeding,the rats in the model group underwent induction with a high-fat diet combined with streptozotocin(STZ)injection to establish the diabetes model.Fasting blood glucose≥16.7 mmol/L after modeling was regar-ded as the criterion for successful model establishment.The successfully modeled rats were further divided into five groups by the random number table method,with 10 rats in each group:a model group,a high-dose Huan-glian Jiedu Decoction group(6.30 g·kg-1),a medium-dose Huanglian Jiedu Decoction group(3.15 g·kg-1),a low-dose Huanglian Jiedu Decoction group(1.58 g·kg-1),and a control group(metformin,200 mg·kg-1).From the second day after modeling,rats in each Huanglian Jiedu Decoction group and the control group were given the corresponding drugs by intragastric administration for consecutive 21 days.Rats in the blank group and the model group were given an equal volume of normal saline by intragastric administration synchro-nously.After the completion of drug intervention,all rats were fasted for 12 hours with free access to drinking water.Blood samples were collected via tail vein puncture to detect fasting blood glucose.Subsequently,fresh feces were collected for DNA extraction and qPCR detection.Results The abundance of Lactobacillus in the model group was significantly lower than that in the blank group.After intervention with Huanglian Jiedu Decoc-tion and metformin,the abundance of Lactobacillus was markedly restored.Compared with the model group,both the low dose Huanglian Jiedu Decoction group and the control group showed a significant increase in Lacto-bacillus abundance(P<0.05).Conclusion Huanglian Jiedu Decoction can effectively increase the intestinal abundance of Lactobacillus in diabetic rats,and the low-dose intervention exhibits the most prominent therapeu-tic effect.
