消肿止痛合剂对大鼠骨骼肌损伤后MyoD蛋白及mRNA表达的影响OA
Effects of Xiaozhong Zhitong Mixture on the Expressions of MyoD Protein and mRNA in Rats after Skeletal Muscle Injury
目的:基于骨骼肌损伤后模型大鼠骨骼肌组织生肌决定因子(myogenic determination fac-tor,MyoD)蛋白及mRNA的表达,探讨消肿止痛合剂对损伤后的骨骼肌修复再生的影响.方法:将80只实验大鼠通过随机数字表法分为空白组、对照组、模型组及治疗组,每组20只,每组大鼠再随机分为1天、7天、14天及21天组,每组5只.空白组予生理盐水灌胃;对照组予布洛芬溶液灌胃;模型组予生理盐水灌胃;治疗组按10 mL·kg-1剂量灌胃消肿止痛合剂.干预结束后,利用RT-PCR法检测MyoD mRNA表达水平,蛋白免疫印迹法检测MyoD蛋白表达水平.结果:与空白组比较,第1天模型组骨骼肌中MyoD蛋白及mRNA表达无明显差异(P>0.05),第7、14、21天模型组骨骼肌中MyoD蛋白及mRNA表达随着时间延长,逐渐上调(P<0.05);与模型组比较,第7、14天治疗组骨骼肌中MyoD蛋白及mRNA表达上调(P<0.05).结论:消肿止痛合剂能够通过上调MyoD蛋白及mRNA表达,促进骨骼肌损伤后的修复及再生.
Objective:To investigate the effect of Xiaozhong Zhitong Mixture on the repair and regeneration of injured skeletal muscle based on the expression of myogenic determination factor(MyoD)protein and mRNA in the skeletal muscle tissue of rat models after skeletal muscle injury.Methods:A total of 80 experimental rats were randomly divided into a blank group,a control group,a model group,and a treatment group using a random number table method,with 20 rats in each group.Rats in each group were further randomly subdivided into four subgroups according to the time points of day 1,day 7,day 14,and day 21,with 5 rats in each subgroup.The blank group received normal saline by gavage;the control group received ibuprofen solution by gavage;the model group received normal saline by gavage;and the treatment group received Xiaozhong Zhitong Mixture at a dose of 10 mL/kg by gavage.After the intervention,the expression level of MyoD mRNA was detected by RT-PCR,and the expression level of MyoD protein was detected by Western blotting.Results:Compared with the blank group,there was no significant difference in the expression levels of MyoD protein and mRNA in the skeletal muscle of the model group on day 1(P>0.05).However,on days 7,14,and 21,the expression levels of MyoD protein and mRNA in the skeletal muscle of the model group were gradually up-regulated over time(P<0.05).Compared with the model group,the expression levels of MyoD protein and mRNA in the skeletal muscle of the treatment group were significantly up-regulated on days 7 and 14(P<0.05).Conclusion:Xiaozhong Zhitong Mixture can promote the repair and regeneration of injured skeletal muscle by up-regulating the expression of MyoD protein and mRNA.
