乳酸化是一种新型代谢依赖性的翻译后修饰,在胃癌的发生发展及治疗耐药形成中发挥关键作用.乳酸化不仅通过调控DNA损伤修复、细胞周期、凋亡途径及肿瘤代谢微环境促进肿瘤细胞增殖和转移,还通过增强免疫抑制、加速药物清除降低治疗效果.目前,抑制乳酸生成、阻断乳酸转运、调控乳酸化修饰酶系及其下游效应分子等是靶向胃癌乳酸化的主要手段,已在细胞和动物实验中展现出良好应用前景.本研究将系统阐述乳酸化诱导胃癌发生和耐药的机制及其相应干预策略,旨在为胃癌的生物标志物诊断和精准化治疗提供新思路.

Lactylation is a novel metabolism-dependent post-translational modification that plays a crucial role in the initiation,pro-gression,and therapeutic resistance of gastric cancer.Lactylation not only promotes tumor cell proliferation and metastasis by regulating DNA damage repair,cell cycle progression,apoptotic pathways,and the metabolic tumor microenvironment,but also reduces treatment efficacy by en-hancing immuno suppression and accelerating drug clearance.Currently,major strategies for targeting lactylation in gastric cancer include in-hibiting lactate production,blocking lactate transport,and modulating lactylation-related enzymes and their downstream effectors.The aim of this study is to integrate the existing gastric cancer treatment methods that mainly focus on interfering with the lactate process.The main signifi-cance lies in attempting to promote the transformation of relevant biomarkers into clinical applications,with the expectation of enhancing the precision treatment level for patients with gastric cancer.
