内质网应激与视网膜疾病的研究进展OA
The research progress of endoplasmic reticulum stress and retinal diseases
视网膜组织是形成正常视觉的重要组织,维持正常视觉功能需要合成大量蛋白质;内质网是蛋白质和脂质合成的核心细胞器,主要承担细胞内蛋白的合成、折叠、翻译、组装、修饰和转运作用.在蛋白质折叠的过程中,会受Ca2+浓度失调、缺血缺氧、血糖异常、基因突变等因素影响导致出错,从而引发内质网应激;进一步通过启动未折叠蛋白反应、内质网超载反应以及固醇调节元件结合蛋白通路诱导炎症、氧化应激以及细胞凋亡机制,参与多种视网膜疾病的病理过程.本文就内质网应激与常见视网膜疾病的关联及治疗干预进展进行综述.
The retina is indispensable for normal visual function,which relies on massive protein synthesis.As the core organelle gov-erning protein and lipid biosynthesis,the endoplasmic reticulum is primarily responsible for intracellular protein synthesis,folding,translation,assembly,modification and trafficking.Disruptions including Ca2+homeostasis imbalance,ischemia-hypoxia,abnormal blood glucose and gene mutations can induce protein misfolding,thus triggering endoplasmic reticulum stress.Furthermore,endoplasmic reticulum stress mediates in-flammatory response,oxidative stress and apoptosis via activating the unfolded protein response,endoplasmic reticulum overload response and sterol regulatory element-binding protein pathway,contributing to the pathogenesis of multiple retinal diseases.This review summarizes the cor-relation between endoplasmic reticulum stress and common retinal diseases,as well as recent advances in targeted therapeutic interventions.
