天然成分及多因子靶向PI3K/AKT/mTOR通路在口腔鳞状细胞癌中的研究进展OA
Research progress on natural compounds and multi-agent targeting of the PI3K/AKT/mTOR Pathway in oral squamous cell car-cinoma
口腔鳞状细胞癌(OSCC)的高侵袭性与治疗耐药性导致临床预后不良,靶向 PI3K/AKT/mTOR信号通路成为突破其治疗瓶颈的关键策略之一.该通路异常激活通过调控细胞增殖、代谢及转移驱动OSCC进展.本综述系统总结天然活性成分及多因子靶向抑制该通路的分子机制,涵盖凋亡诱导、自噬促进、糖酵解抑制及免疫逃逸逆转等作用,并揭示其增敏放化疗的协同潜力,旨在为开发低毒、精准的OSCC靶向治疗提供理论及转化依据.
The high invasiveness and therapeutic resistance of oral squamous cell carcinoma(OSCC)contribute to its poor clinical prog-nosis,rendering the targeting of the PI3K/AKT/mTOR signaling pathway a pivotal strategy to overcome therapeutic bottlenecks.Aberrant activa-tion of this pathway drives OSCC progression by modulating cell proliferation,metabolism,and metastasis.This review systematically summa-rizes the molecular mechanisms underlying the targeted inhibition of the PI3K/AKT/mTOR pathway by natural bioactive components and multi-functional factors,encompassing apoptosis induction,autophagy promotion,glycolysis suppression,and reversal of immune evasion.Additional-ly,it highlights their synergistic potential in sensitizing radiotherapy and chemotherapy.These findings provide a theoretical and translational foundation for developing low-toxicity,precision-targeted therapeutic strategies against OSCC.
