目的:探讨糖尿病肾病(DN)大鼠血清糖基化终末产物(AGEs)与肾小球足细胞裂孔膜蛋白 Nephrin、Podocin 表达的相关性,以阐明AGEs在DN足细胞损伤中的作用机制.方法:将24只Wistar大鼠随机分为对照组和实验组,实验组通过高脂高糖饲料喂养联合链脲佐菌素(STZ)诱导DN模型.采用ELISA法检测血清AGEs水平,Western blot和免疫组化法检测肾组织中Nephrin、Podocin的表达,并通过Pearson相关性分析评估AGEs与足细胞蛋白的关系.结果:实验组血清AGEs水平为(356.82±42.57)ng/mL,高于对照组的(128.35±21.64)ng/mL,差异有统计学意义(P<0.01),且实验组 Nephrin、Podocin 表达降低(P<0.01).相关性分析显示,血清 AGEs水平与Nephrin(r=-0.655)和Podocin(r=-0.639)表达呈负相关(P<0.01).结论:DN大鼠血清AGEs水平升高与足细胞裂孔膜蛋白Nephrin、Podocin表达降低密切相关,提示AGEs可能通过损伤足细胞结构完整性参与DN的发病机制,为DN的早期干预提供了潜在靶点.

Objective:To investigate the correlation between serum glycosylation end products(AGEs)and the expression of glomerular podocyte lacunar membrane proteins Nephrin and Podocin in diabetic nephropathy(DN)rats to elucidate the mechanism of AGEs in DN podocyte injury.Methods:Twenty-four Wistar rats were randomly divided into a control group and an experimental group,and the experimental group was fed by high-fat and high-sugar chow combined with streptozotocin(STZ)to induce DN model.The levels of serum AGEs were detected by ELISA,and the expressions of Nephrin and Podocin in renal tissues were detected by Western blot and immunohistochemistry,and the relation-ship between AGEs and podocyte proteins was evaluated by Pearson correlation analysis.Results:The level of serum AGEs in the experimental group(356.82±42.57)ng/mL was significantly higher than that in the control group(128.35±21.64)ng/mL(P<0.01),and the expression of Nephrin and Podocin was significantly lower(P<0.01).Correlation analysis showed that serum AGEs levels were significantly negatively correlated with Nephrin(r=-0.655)and Podocin(r=-0.639)expression(P<0.01).Conclusion:Elevated levels of serum AGEs in DN rats were closely correlated with decreased expression of podocyte injury lacunar membrane proteins Nephrin and Podocin,suggesting that AGEs may be involved in the pathogenesis of DN by impairing the structural integrity of the podocyte injury,and providing a potential target for early intervention in DN.
