基于牛血清蛋白修饰的超顺磁性氧化铁双模态结直肠癌分子探针的制备及验证OA
Preparation and validation of bimodal molecular probe for colorectal cancer based on bovine serum albumin-modified superparamagnetic iron oxide
目的 制备一种细胞靶向超顺磁性氧化铁荧光-磁共振双信号分子探针Cy5.5-antiGlut1-BSA⋅SPIONs,评估其基本性质并探究细胞荧光及磁共振成像能力.方法 采用高温热注射法合成油溶性SPIONs;采用超声乳化法合成水溶性BSA⋅SPIONs探针前体;与抗肿瘤多克隆抗体(Glut1)和红外荧光素Cy5.5偶联后,制备出Cy5.5-antiGlut1-BSA⋅SPIONs靶向双信号分子探针.对Cy5.5-antiGlut1-BSA⋅SPIONs探针的形貌、粒径、光学性质、弛豫率等关键性质进行基本表征,并创新性地使用Cy5.5-antiGlut1-BSA⋅SPIONs分子探针在荧光共聚焦显微镜下观察Glut1在组织活检中肿瘤部位的表达和分布.结果 此分子探针直径为20~40 nm,在酸碱性条件下均具有胶体稳定性和生物相容性.它表现出强荧光性和高弛豫性(r2:每mmol/L Fe3+为129.92 s-1).随着探针浓度的增加磁共振成像T2信号逐渐增强.通过细胞毒性和组织学毒性检测,该分子探针基本无毒性.结论 Cy5.5-antiGlut1-BSA⋅SPIONs分子探针具有优良的荧光-磁共振双模态成像能力,对结直肠癌细胞HCT116具有特异靶向性,是用于结直肠癌细胞分子诊断和体外活检分析的有前途分子探针.
Objective To developed a cell-targeted superparamagnetic iron oxide fluorescence-magnetic resonance dual-signal molecular probe,Cy5.5-antiGlut1-BSA⋅SPIONs was prepared.To evaluate its basic properties and explore cell fluorescence and magnetic resonance imaging capabilities.Methods Oil-soluble SPIONs were synthesized by high-temperature hot injection;The water-soluble BSA⋅SPIONs probe precursor was synthesized by phacoemulsification;After coupling with the anti-tumor polyclonal antibody(Glut1)and the infrared fluorescein Cy5.5,the Cy5.5-antiGlut1-BSA SPIONs targeted dual-signal molecular probe was prepared.The key properties such as morphology,particle size,optical properties and relaxation rate of the Cy5.5-antiGlut1-BSA·SPIONs probe were basically characterized.Furthermore,the Cy5.5-antiGlut1-BSA·SPIONs molecular probe was innovatively used to observe the expression and distribution of Glut1 at the tumor site in tissue biopsy under a fluorescence confocal microscope.Results This diameter of this molecular probe was 20-40 nm and it exhibited colloidal stability and biocompatibility under both acidic and alkaline conditions.It exhibited strong fluorescence and high relaxation(r2:129.92 s-1 per mmol/L Fe3+).The T2 signal of magnetic resonance imaging gradually increased with the increase of probe concentration.Through cytotoxicity and histological toxicity tests,this molecular probe showed non-toxic.Conclusion The Cy5.5-antiGlut1-BSA·SPIONs molecular probe possessed excellent fluorescence-magnetic resonance dual-mode imaging capabilities and specific targeting ability for colorectal cancer cells HCT116.It was a promising molecular probe for in vivo MRI-fluorescent diagnosis and ex vivo biopsy analysis of colorectal cancer cells.
