[背景]河豚毒素(TTX)是一种结构独特、毒性强烈的天然弱碱性毒素.它通过特异性阻断电压门控钠离子通道,引发神经麻痹并产生强大的中枢镇痛效果,因显著的药理学价值,其已成为天然产物研究的热点.TTX 尽管潜力巨大,但实际应用仍然受制备工艺复杂和安全窗口狭窄等缺点的制约.[目的]本文旨在从"化毒为药"的转化医学视角,系统阐明 TTX 提取、纯化、医学药理活性与安全之间的关系,并为突破其在渔业领域的研究瓶颈提供策略参考.[进展]本文综述了 TTX 在提取、纯化及医学药理活性研究方面的关键进展.TTX 提取方法由传统恒温水浴法向超声辅助提取法发展,纯化技术则由柱层析和膜分离法向高分辨液相色谱-质谱联用(HPLC-MS)技术和免疫磁珠体系演进,显著提升了TTX 的纯度和回收率.医学药理活性研究表明,TTX 通过阻断钠离子通道在镇痛、抗心律失常、抑制肿瘤转移及局部麻醉等方面具有潜在的应用价值.近年来,脂质体、纳米颗粒等药物递送系统被用于 TTX 的包载与控释,可有效降低 TTX 毒性并延长药效,为其安全药用化提供了新思路.[结论]TTX 具有巨大的药用潜力,但提取和纯化的困难使其"难量产",安全窗口的狭窄使其"难应用".未来需探究低成本化、产业化提取和纯化工艺,开发靶向给药系统,从而逐步实现 TTX 的高值化应用以及"渔业资源-临床应用"的跨学科结合.

[Background]Tetrodotoxin(TTX)is a natural weak alkaline toxin with a unique structure and strong toxicity.It causes nerve palsy and produces a strong central analgesic effect by specifically blocking voltage-gated sodium channels.Because of its significant pharmacological value,it has become a hot spot in natural product research.Although its potential is huge,its practical application is still restricted by its complex preparation process and narrow safety window.[Objective]This paper aims to systematically clarify the rela-tionship among TTX extraction,purification,medical pharmacological activities and safety from the perspect-ive of translational medicine,and to provide a strategic reference for breaking through the bottleneck of TTX re-search in the field of fisheries.[Progress]In this paper,the key progress of TTX in extraction,purification and functional activity research is reviewed.TTX extraction has developed from the traditional constant temperat-ure water bath method to ultrasonic-assisted extraction,and purification technology has evolved from column chromatography and membrane separation to high-resolution high-performance liquid chromatography-mass spectrometry(HPLC-MS)and immunomagnetic bead systems,which has significantly improved the purity and recovery rate of TTX.Functional activity studies have shown that TTX has potential application value in anal-gesia,anti-arrhythmia,inhibition of tumor metastasis and local anesthesia by blocking sodium ion channels.In recent years,drug delivery systems such as liposomes and nanoparticles have been used for the encapsulation and controlled release of TTX,which can effectively reduce toxicity and prolong efficacy,providing a new idea for its safe medicinal use.[Conclusion]TTX has great medicinal potential,but the difficulty of extraction and purification makes it"difficult to produce",and the narrow safety window makes it"difficult to apply".In the future,it will be necessary to explore low-cost,industrialized extraction and purification processes,and to devel-op targeted drug delivery systems,so as to gradually realize the high-value application of TTX and the interdis-ciplinary combination of"fishery resources-clinical application".
