Huaiqihuang Granules Inhibit Inflammation and Proliferation in Psoriasis:Mechanisms Based on Network Pharmacology and in Vivo Experimental Validation

目的:基于网络药理学合并动物试验探究槐杞黄颗粒对寻常型银屑病的干预机制.方法:利用网络药理学预测槐杞黄颗粒对银屑病干预机制,通过HERB数据库、Swiss Target Prediction数据库筛选活性成分靶点.通过GeneCard、Disgent数据库获取寻常型银屑病相关基因.通过Cytoscape软件构建槐杞黄颗粒成分-靶点-银屑病网络,利用STRING数据库绘制蛋白相互作用(PPI)网络图,分析槐杞黄颗粒治疗寻常型银屑病的核心靶点.通过DAVID数据库进行基因本体(GO)功能注释和京都基因与基因组百科全书(KEGG)通路富集分析,预测其富集的信号通路并进行可视化展示.选择60只KM雄性小鼠,随机分为空白组、模型组、银屑胶囊组(0.6 g/kg)、槐杞黄高剂量组(8 g/kg)、槐杞黄中剂量组(4 g/kg)、槐杞黄低剂量组(2 g/kg),各10只.采用5%咪喹莫特乳膏涂抹小鼠背部裸露皮肤建立银屑病动物模型,治疗1周后,对小鼠皮损处银屑病面积和严重程度指数(psoriasis area and severity index,PASI)评分,记录小鼠耳廓肿胀度、耳廓肿胀率,甲苯胺蓝染色观察小鼠背部皮损组织肥大细胞数,ELISA检测小鼠血清IL-17、TNF-α表达水平,免疫组化检测各组小鼠皮损处VEGF、Ki67表达.结果:经过检索及筛选共得到槐杞黄颗粒药物活性成分作用靶点1 016个,银屑病作用靶点1 169个,药物疾病共同靶点190个.槐杞黄颗粒中槐耳、枸杞、黄精与银屑病分别有49个、179、133个交集基因;对槐杞黄颗粒的核心靶点进行GO功能富集分析,共得到2 639个生物过程、85种细胞组分、205种分子功能,核心靶点主要与炎症反应调控及细胞凋亡、增殖调控的生物过程相关.与模型组比较,槐杞黄高剂量组小鼠PASI评分、耳廓肿胀度、耳廓肿胀率、背部皮肤肥大细胞数、血清IL-17、TNF-α水平、皮肤VEGF、Ki67表达水平均有不同程度的降低(P<0.01).结论:槐杞黄颗粒可通过抑制炎症反应、调控细胞凋亡与增殖发挥控制银屑病的作用.

Objective:To investigate the intervention mechanism of Huaiqihuang Granules in psoriasis vulgaris based on network phar-macology and animal experiments.Methods:Network pharmacology was used to predict the intervention mechanism of Huaiqihuang Granules in psoriasis.Active ingredient targets were screened using the HERB database and Swiss Target Prediction database.Genes related to psoriasis vulgaris were obtained from the GeneCard and DisGeNET databases.A Huaiqihuang Granules component-target-psoriasis network was constructed using Cytoscape software,and a protein-protein interaction(PPI)network diagram was drawn using the STRING database to analyze the core targets of Huaiqihuang Granules in treating psoriasis vulgaris.Gene Ontology(GO)function-al annotation and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were performed using the DAVID database to predict enriched signaling pathways,and the results were visually presented.Animal experiments:Sixty KM male mice were randomly divided into a blank group,model group,Yinxie Capsule group(0.6 g/kg),Huaiqihuang high-dose group(8 g/kg),Huaiqihuang medium-dose group(4 g/kg),and Huaiqihuang low-dose group(2 g/kg),with 10 mice in each group.A psoriasis ani-mal model was established by applying 5%imiquimod cream to the shaved back skin of the mice.After one week of treatment,the Psoriasis Area and Severity Index(PASI)score of the mouse skin lesions was assessed,and the degree of ear swelling and ear swelling rate were recorded.Toluidine blue staining was used to observe the number of mast cells in the back skin lesion tissue of the mice.Ser-um levels of IL-17 and TNF-α were detected by ELISA,and the expression of VEGF and Ki67 in the skin lesions of mice in each group was detected by immunohistochemistry.Results:A total of 1,016 drug-active ingredient targets and 1,169 psoriasis-related targets were identified after retrieval and screening,with 190 common targets between the drug and the disease.Poria cocos,Lycium barbarum,and Polygonatum sibiricum in Huaiqihuang Granules had 49,179,and 133 overlapping genes with psoriasis,respectively.GO func-tional enrichment analysis of the core targets of Huaiqihuang Granules yielded 2,639 biological processes,85 cellular components,and 205 molecular functions.The core targets were mainly associated with biological processes such as the regulation of inflammatory re-sponse and the regulation of cell apoptosis and proliferation.Compared with the model group,the Huaiqihuang high-dose group showed varying degrees of reduction in PASI score,degree of ear swelling,ear swelling rate,number of mast cells in the back skin,serum lev-els of IL-17 and TNF-α,and expression levels of VEGF and Ki 67 in the skin(P<0.01).Conclusion:Huaiqihuang Granules can exert a therapeutic effect on psoriasis by inhibiting the inflammatory response and regulating cell apoptosis and proliferation.
