目的 基于"阳化气,阴成形"中医理论,结合细胞器互作调控自噬的现代研究,基于计算仿真探讨人参皂苷-莪术醇配伍影响线粒体-溶酶体互作调控卵巢癌细胞自噬的作用机制.方法 构建"中药干预-细胞器互作-卵巢癌细胞自噬"(TCMI-OI-OCCA)多主体模型,利用NetLogo软件仿真系统动态.通过行为空间实验获取不同参数组合下的系统行为数据,并采用随机森林回归模型分析关键变量对系统平衡的影响重要性.结果 随机森林回归模型拟合优度高(R² ≈ 0.999),变量重要性分析显示,癌细胞增殖概率(cellp)、仿真时间(time)与自噬概率(cella)是影响系统平衡的最关键预测因子;人参皂苷与莪术醇的干预参数(gin,gine,cur,cure)亦显示出稳定的重要性,提示其在调节系统动态平衡中具有明确作用;线粒体-溶酶体正负向互作参数(pi,ni)在当前模型中对预测精度贡献不显著.结论 人参皂苷-莪术醇配伍可能通过调节线粒体-溶酶体互作网络,影响卵巢癌细胞自噬与增殖的动态平衡,体现了"温阳化瘀"治法在系统层面对"阳化气-阴成形"平衡的调控作用.

Objective Based on the traditional Chinese medicine theory of"Yang Transforms Qi,Yin Constitutes Form"and incorporating modern research on organelle interactions in autophagy regulation,this study aimed to computationally investigate the mechanism by which ginsenoside-curcumol combination regulates autophagy in ovarian cancer cells via influencing mitochondria-lysosome interactions.Methods A multi-agent model called"Traditional Chinese Medicine Intervention-Organelle Interaction-Ovarian Cancer Cell Autophagy"(TCMI-OI-OCCA)was constructed and simulated using NetLogo software to explore the system dynamics.Behavioral space experiments were conducted to obtain system behavior data under different parameter combinations.A random forest regression model was employed to analyze the importance of key variables on the system equilibrium.Results The random forest regression model demonstrated high goodness-of-fit(R² ≈ 0.999).Variable importance analysis revealed that cancer cell proliferation probability(cellp),simulation time(time),and autophagy probability(cella)were the most critical predictors influencing system equilibrium.The intervention parameters for ginsenoside and curcumol(gin,gine,cur,cure)also showed stable importance,indicating their definite role in modulating the system's dynamic balance.The mitochondria-lysosome positive and negative interaction parameters(pi,ni)did not contribute significantly to prediction accuracy in the current model.Conclusion The ginsenoside-curcumol combination may regulate the dynamic balance between autophagy and proliferation in ovarian cancer cells by modulating the mitochondria-lysosome interaction network.This effect embodies how the"Warming Yang and Resolving Stasis"therapeutic principle systemically regulates the balance of"Yang Transforms Qi,Yin Constitutes Form".
