补肺益肾方对慢性阻塞性肺疾病大鼠尿液代谢谱的影响OA
Effect of Bufei Yishen Formula on Urinary Metabolic Profiling in Rats with Chronic Ob-structive Pulmonary Disease
目的:基于超高效液相色谱-四极杆-静电场轨道阱串联质谱法(UHPLC-QE-MS)的尿液代谢组学方法探究补肺益肾方对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)大鼠尿液代谢产物的影响.方法:将24 只SD 大鼠随机分成正常对照、模型对照组、补肺益肾方 3.7 g/kg 组、羧甲司坦 0.14 g/kg 组,观察大鼠的体质量、肺指数、肺湿干质量比变化;检测大鼠肺功能;观察肺外观、组织病理及超微结构;UHPLC-QE-MS 法分析尿液代谢轮廓;多元统计方法分析差异代谢物;MetaboAnalyst 数据库分析差异代谢物的富集通路.结果:与正常对照组相比,模型对照组大鼠肺指数、肺湿干质量比显著升高(P<0.01),肺功能 FVC、FEV0.1 及 FEV0.1/FVC 显著降低(P<0.01),肉眼见部分肺叶肿大发白,病理染色显示肺泡壁断裂、融合及炎细胞浸润,超微结构显示肺泡Ⅱ型上皮细胞板层小体空泡化,线粒体嵴肿胀明显,胞内见脂滴;与模型对照组相比,补肺益肾方3.7 g/kg 组 COPD 大鼠肺指数、肺湿干质量比明显降低(P<0.05 或P<0.01),肺功能显著增强(P<0.01),肺外观及病理损伤改善.尿液代谢组学分析筛选模型对照组潜在的生物标志物共180 个,补肺益肾方3.7 g/kg 组对其中的106 个代谢物呈现良好的回调趋势,主要富集到苯丙氨酸代谢、Vit B6 代谢、乙醛酸和二羧酸代谢、精氨酸和脯氨酸代谢、丙氨酸、天门冬氨酸和谷氨酸代谢、烟酸和烟酰胺代谢、色氨酸代谢、柠檬酸循环及精氨酸的生物合成等多个通路.结论:补肺益肾方对 COPD 具有缓解作用,可能与其改善内源性代谢物紊乱,恢复正常机体代谢活动有关.
Objective:This study aims to investigate the effects of Bufei Yishen Formula(补肺益肾方,BYF)on u-rinary metabolites in rats with chronic obstructive pulmonary disease(COPD)based on urine metabolomics using ultra-high-performance liquid chromatography-quadrupole-electrostatic field orbitrap mass spectrometry(UHPLC-QE-MS).Methods:24 SD rats were randomly divided into a normal control group,a model control group,a BYF 3.7 g/kg group,and an S-carboxymethyloysteine(S-CMC)0.14 g/kg group.Body weight,lung index,and lung wet/dry weight ratio were observed.Pulmonary function,gross lung appearance,histopathology,and ul-trastructure were detected.Urinary metabolic profiles were analyzed by UHPLC-QE-MS.Dif-ferential metabolites were analyzed by a multivariate statistical method,and enriched metabolic pathways were analyzed by the MetaboAnalyst database.Results:Compared with the normal control group,rats in the model control group showed increased lung index and lung we/dry weight ratio(P<0.01),and decreased lung function indexes including forced vital capacity(FVC),forced expiratory volume in 0.1 sec-ond(FEV0.1),and FEV0.1/FVC(P<0.01).Gross lung observations revealed enlargement and pallor of some lung lobes.Histopathological staining showed alveolar wall rupture,fusion,and inflammatory cell infiltration.Ultrastructure a-nalysis showed vacuoliza-tion of lamellar bodies in alveolar type II epithelial cells,mitochondrial cristae swelling,and in-tracellular appearance of lipid droplets.Compared with the model control group,rats in the BYF 3.7 g/kg group exhibi-ted partially restored body weight,reduced lung index and wet/dry weight ratio(P<0.05),improved pulmonary function(P<0.01),and alleviated macroscopic lung injury.A total of 180 potential urinary biomarkers were identified in the model control group,among which 106 metabolites showed a clear trend of regulation after BYF 3.7 g/kg intervention.These metabolites were mainly enriched in pathways including phenylalanine metabolism,vitamin B6 metabolism,glyoxy-late and dicarboxylate metabolism,arginine and proline metabolism,alanine,aspartate and glutamate metabolism,nicoti-nate and nicotinamide metabolism,tryptophan metabo-lism,citrate cycle,and arginine biosynthesis.Conclusions:BYF may alleviate COPD by im-proving endogenous metabolic disturbances and restoring normal metabolic activity.
