目的 探讨芪七连方治疗高血压的分子机制.方法 以超高效液相色谱-串联质谱(UHPLC-MS/MS)测定芪七连方的成分;以 SWISS ADME 和 SWISS Target Prediction 数据库预测其活性成分及靶点;从 DisGeNET、DrugBank、GeneCards、OMIM、TTD 数据库获取高血压病基因靶点;将活性成分靶点与高血压病基因靶点取交集;将交集靶点导入 STRING 数据库预测蛋白质-蛋白质相互作用(PPI)网络,再使用 Cytoscape 3.9.1 的cytoHubba 插件 MCC 模式取前 5 位作为核心基因;用 Metascape 数据库进行 GO 分析和 KEGG 通路富集分析;选取核心活性成分与核心基因进行分子对接验证结合活性.结果 共获得芪七连方 65 种活性成分和 803 个靶点,1 304 个高血压基因靶点.芪七连方的核心成分为木兰花碱、柚皮素、槲皮素-3-甲基醚、异鼠李素,其治疗高血压的核心靶点是 STAT3、PTPN11、JAK1 等;芪七连方通过脂质与动脉粥样硬化、PI3K/Akt 信号通路、AGE-RAGE 信号通路等信号通路调控高血压,分子对接结果显示核心成分和核心靶点的结合能≤-5.0 kcal·mol-1,表明结合效果良好.结论 芪七连方通过多成分、多靶点、多通路防治高血压.

Objective To investigate the molecular mechanism of Qiqilian Formula in treating hypertension.Methods UHPLC-MS/MS was employed to identify the components of Qiqilian Formula.Active components and their corresponding targets were predicted using the SWISS ADME and SWISS Target Prediction databases.Hypertension-related genes were obtained from the DisGeNET,DrugBank,GeneCards,OMIM,and TTD databases.Intersecting targets were derived by matching the predicted active component targets with hypertension-related gene targets.These intersecting targets were imported into the STRING database to predict the protein-protein interaction(PPI)network,and the cytoHubba plugin(MCC mode)in Cytoscape 3.9.1 was used to select the top 5 genes as core genes.GO and KEGG pathway enrichment analyses were performed using the Metascape database.Molecular docking was conducted between the core active components and core genes.Results A total of 65 active components and 803 corresponding targets were identified for Qiqilian Formula,along with 1 304 hypertension-related gene targets.The core active components of Qiqilian Formula were magnoflorine,naringenin,quercetin-3-methyl ether,and isorhamnetin.The core targets for treating hypertension included STAT3,PTPN11,and JAK1.Qiqilian Formula appears to regulate hypertension through pathways such as lipid and atherosclerosis,the PI3K/Akt signaling pathway,and the AGE-RAGE signaling pathway.Molecular docking results indicated favorable binding interactions between the core components and core targets,with binding energies≤-5.0 kcal·mol-1.Conclusion Qiqilian Formula exerts its preventive and therapeutic effects on hypertension through a multi-component,multi-target,and multi-pathway mechanism.
