多重RT-RPA技术检测常见儿童腹泻病原体的方法研究OA
Research on the method of multiplex RT-RPA technology for detecting common pathogens causing diarrhea in children
目的 建立一种可同时检测多种常见儿童腹泻病原体的反转录重组酶聚合酶扩增(reverse transcription recombinase polymerase amplification,RT-RPA)方法.方法 选取轮状病毒、腺病毒、诺如病毒基因组的保守序列设计重组酶聚合酶扩增(recombinase polymerase amplification,RPA)引物探针,建立多重RT-RPA检测体系,并对建立的多重RT-RPA反应体系的敏感度、特异性、重复性进行验证,同时对建立的体系进行小样本临床验证.结果 本研究建立的多重RT-RPA对轮状病毒、腺病毒、诺如病毒的检测敏感度分别可达101copies/μl、102copies/μl、102copies/μl,且不与其他病毒发生交叉反应,重复检测不同浓度的样本10次,Ct值的标准差变异系数均小于5%,临床样本常见儿童腹泻病原体的检出率与PCR法和胶体金法无明显差异.结论 本研究建立的多重RT-RPA具有较好的敏感度、特异性和重复性,可为儿童腹泻病原体的检测提供新的思路.
Objective To establish a reverse transcription recombinase polymerase amplification(RT-RPA)that can simultaneously detect multiple common pathogens causing diarrhea in children.Methods Conservative sequences of rotavirus,adenovirus,and norovirus genomes were selected to design recombinase polymerase amplification(RPA)primer and probe,and a multiplex RT-RPA detection system was established.The sensitivity,specificity,and repeatability of the established multiplex RT-RPA reaction system were validated,and small clinical sample validation was conducted on the established system.Results The multiplex RT-RPA established in this study has a detection sensitivity of 101copies/μl,102copies/μl and 102copies/μl for rotavirus,adenovirus,and norovirus,respectively,and there was no cross-reaction with other common viruses.Different concentrations of samples were tested 10 times,and the standard deviation of Ct values was small,with coefficients of variation less than 5%.There is no significant difference in the detection rate of common pediatric diarrhea pathogens in clinical samples between PCR method and the colloidal gold method.Conclusion The multiplex RT-RPA established in this study has good sensitivity,specificity,and reproducibility,and can provide new ideas for the detection of pathogens in childhood diarrhea.
