Objective Through systematic evaluation and Meta-analysis,a comprehensive quantitative assessment was conducted on the association between oral contraceptive(OCs)use and the risk of depressive symptoms in multiple clinical trials.Methods Literature search was conducted to identify studies on the impact of OCs on depression across multiple databases,ultimately incorporating 14 clinical trials.Random-effects Meta-analysis was performed to combine risk ratios,with subgroup analyses conducted based on study sources and depression symptom evaluations to explore heterogeneity sources.Results Compared with control group,the use of OCs did not significantly increase the risk of depressive symptoms(RR=1.09,95%CI:0.83-1.42,P=0.540).Significant heterogeneity was observed across studies,with subgroup analyses indicating that the impact of OCs on depression risk may vary across different countries,study methods,depression assessment tools,types of OCs,and age groups.Further Meta-regression analysis revealed that OCs type,mean age,and follow-up duration were also significant sources of heterogeneity.Conclusion Current clinical trial evidence does not indicate that OCs significantly cause or exacerbate depressive symptoms in adult women.However,this does not imply that OCs have no effect on depressive symptoms.Heterogeneity analysis suggests that their impact may vary across different countries,with differences in study methodologies and depression assessment tools.
