扶正祛瘀方通过抑制NLRP3炎性小体激活治疗慢性子宫内膜炎的实验研究OA
Study on the Treatment of Chronic Endometritis with Fuzheng Quyu Formula by Inhibiting NLRP3 Inflammasome Activation
目的:探讨扶正祛瘀方对慢性子宫内膜炎(chronic endometritis,CE)的治疗作用及潜在机制,验证其是否通过清除活性氧(ROS)抑制NOD样受体热蛋白结构域相关蛋白3(NLRP3)炎性小体激活.方法:采用宫腔注射苯酚胶浆构建CE大鼠模型,将24只Wistar大鼠随机分为对照组、模型组、抗生素组[18mg(kg·d)]及扶正祛瘀方组[4.4g(kg·d)].治疗干预后通过苏木精和伊红(HE)染色观察子宫内膜病理变化;酶联免疫吸附试验(ELISA)检测血清炎症因子水平;免疫荧光检测子宫内膜ROS含量;Western blot分析NLRP3炎性小体相关蛋白[NLRP3、凋亡相关颗粒样蛋白(ASC)、半胱天冬酶-1(caspase-1)]表达.结果:与模型组相比,扶正祛瘀方显著改善子宫内膜水肿、炎症细胞浸润等病理损伤,降低血清白细胞介素(IL)-1β、IL-18、肿瘤坏死因子-α(TNF-α)水平,减少子宫内膜ROS蓄积,并抑制NLRP3、ASC和caspase-1蛋白表达,作用效果与多西环素相当.结论:扶正祛瘀方可通过抑制NLRP3炎性小体激活、减轻氧化应激及炎症反应,对慢性子宫内膜炎发挥治疗作用.
Objective:To explore the therapeutic effect and potential mechanism of the Fuzheng Quyu for-mula on chronic endometritis(CE),and to verify whether it inhibits the activation of the NOD-like receptor thermal protein domain associated protein 3(NLRP3)inflammasome by eliminating reactive oxygen species(ROS).Methods:The CE rat model was constructed by intracavitary injection of phenol gelatin.24 Wistar rats were randomly divided into the control group,the model group,the doxycycline group(18 mg/kg/d),and the Fuzheng Quyu formula group(4.4g/kg/d).After the treatment intervention,the pathological changes of the endometrium were observed through hematoxylin and eosin(HE)staining;the levels of inflammatory factors in serum were detected by enzyme-linked immunosorbent assay(ELISA);the content of ROS in the endo-metrium was detected by immunofluorescence;and the expressions of NLRP3 inflammasome-related proteins[NLRP3,apoptosis-associated speck-like protein containing a CARD(ASC),caspase-1]were analyzed by Western blot.Results:Compared with the model group,the Fuzheng Quyu formula significantly improved pathological damages such as endometrial edema and inflammatory cell infiltration,reduced serum levels of interleukin(IL)-1β,IL-18,and tumor necrosis factor-α(TNF-α),decreased endometrial ROS accumulation,and inhibited the expression of NLRP3,ASC,and caspase-1 proteins.The therapeutic effect was comparable to that of doxycycline.Conclusion:The Fuzheng Quyu formula can exert therapeutic effects on CE by inhibit-ing the activation of NLRP3 inflammasome and reducing oxidative stress and inflammatory responses.
