为了建立枸橼酸马罗匹坦原料药的微生物限度检查方法,本试验采用薄膜过滤法进行需氧菌、霉菌和酵母菌总数测定,并对控制菌大肠埃希菌进行检查.结果显示,枸橼酸马罗匹坦具有抑菌作用,薄膜过滤法用pH7.0 无菌氯化钠-蛋白胨缓冲液冲洗滤膜4 次,每次100 mL,可去除该药的抑菌活性,需氧菌、霉菌和酵母菌的回收比值均在0.5~2 范围内,控制菌大肠埃希菌阳性对照组中菌落生长良好,供试液组与阴性对照组中均无菌生长,符合《中国兽药典》要求.结果表明,本研究所建立的薄膜过滤法准确可行,能有效地消除枸橼酸马罗匹坦的抑菌活性,可使用此法进行枸橼酸马罗匹坦原料药的微生物计数与大肠埃希菌控制菌检查,为其质量评价提供了参考和依据.

In order to establish a microbial limit test method for maropitant citrate active pharmaceutical ingredient,this article employed the membrane filtration method to determine the total counts of aerobic bacteria and molds&yeasts,and to detect the specified microorganism Escherichia coli.The results showed that maropitant citrate exhibits antimicrobial activity.Rinsing the filter membrane with pH 7.0 sterile sodium chloride-peptone buffer four times(100 mL,each time)effectively eliminated the inhibitory effect.The recovery rates of aerobic bacteria,molds and yeasts were between 0.5 to 2.In the positive control group for Escherichia coli,the robust colony growth was observed,whereas no colony growth occurred in either the test sample group or the negative control group.The method met the requirements of the Chinese Veterinary Pharmacopoeia.These findings demonstrate that the membrane filtration method established in this study is accurate and feasible;it reliably eliminates the antimicrobial activity of maropitant citrate and is suitable for the microbial enumeration and Escherichia coli detection in maropitant citrate,providing a reference and a solid basis for the quality evaluation.
