天芙止眩方通过调控α7nAChR表达对自发性高血压大鼠下丘脑室旁核小胶质细胞极化的影响OA
Effects of Tianfu Zhixuan Formula on microglial polarization in the hypothalamic paraventricular nucleus of spontaneously hypertensive rats via regulation of α7nAChR expression
目的 探究天芙止眩方对自发性高血压大鼠(SHR)下丘脑室旁核(PVN)小胶质细胞极化的影响.方法 将40 只 SHR 大鼠随机分为模型组、厄贝沙坦组(13.5 mg/kg)和天芙止眩方高、中、低剂量组(10.62、5.31、2.66 g/kg),每组 8 只,连续灌胃给药 8 周,另取 WKY 大鼠作为正常组.采用无创血压仪检测各组大鼠尾动脉收缩压;ELISA 法检测 PVN 中 IL-1β、TNF-α 水平;免疫荧光双标检测PVN 中CD86+/Iba-1+、CD163+/Iba-1+、α7nAChR+/Iba-1+共表达;RT-qPCR 和 Western blot 法检测 PVN 中 CD86、CD163、α7nAChR mRNA 和蛋白表达.利用 LPS 诱导 BV2 细胞,给予不同浓度天芙止眩方含药血清干预,采用 MTT 法检测细胞活力;免疫荧光双标检测细胞 α7nAChR/CD86、α7nAChR/CD163 荧光强度;Western blot 法检测细胞 α7nAChR 蛋白表达.结果 与模型组比较,厄贝沙坦组和天芙止眩方各剂量组大鼠尾动脉收缩压,PVN 中 IL-1β 和 TNF-α 水平、CD86+/Iba-1+表达、CD86 mRNA 和蛋白表达均降低(P<0.05,P<0.01),CD163+/Iba-1+及 α7nAChR+/Iba-1+表达、CD163、α7nAChR mRNA 和蛋白表达均升高(P<0.01).在 LPS 诱导的 BV2 细胞中,天芙止眩方含药血清能提高 α7nAChR 蛋白表达及 α7nAChR、CD163 荧光强度(P<0.05,P<0.01),降低 CD86 荧光强度(P<0.05,P<0.01).结论 天芙止眩方可能通过促进 α7nAChR 表达调控PVN 小胶质细胞极化,减少神经炎症,降低 SHR 大鼠血压.
AIM To explore the effect of Tianfu Zhixuan Formula on microglia polarization in hypothalamic paraventricular nucleus(PVN)of spontaneously hypertensive rats(SHR).METHODS Forty SHR rats were randomly divided into model group,irbesartan group(13.5 mg/kg)and high,middle and low dose groups of Tianfu Zhixuan Formula(10.62,5.31 and 2.66 g/kg),with 8 rats in each group.All rats were administered intragastrically for 8 consecutive weeks.WKY rats were taken as normal group.The systolic blood pressure of tail artery of rats in each group was detected by noninvasive blood pressure meter.The levels of IL-1β and TNF-α in PVN were detected by ELISA.The co-expressions of CD86+/Iba-1+,CD163+/Iba-1+and α7nAChR+/Iba-1+in PVN were detected by immunofluorescence double labeling.RT-qPCR and Western blot were used to detect the mRNA and protein expressions of CD86,CD163 and α7nAChR in PVN.BV2 cells were induced by LPS,intervened by serum containing different concentrations of Tianfu Zhixuan Formula,and the cell viability was detected by MTT assay.The fluorescence intensity of α7nAChR/CD86 and α7nAChR/CD163 cells was detected by immunofluorescence double labeling.The protein expression of α7nAChR in cells was detected by Western blot.RESULTS Compared with the model group,the systolic blood pressure of the caudal artery,the levels of IL-1β and TNF-α in PVN,the expression of CD86+/Iba-1+,and the expression of CD86 mRNA and protein in irbesartan group and Tianfu Zhixuan Formula groups were all decreased(P<0.05,P<0.01),the expressions of CD163+/Iba-1+and α7nAChR+/Iba-1+,and the mRNA and protein expressions of CD163 and α7nAChR increased(P<0.01).In BV2 cells induced by LPS,the serum containing Tianfu Zhixuan Formula could increase the expression of α7nAChR protein and the fluorescence intensity of α7nAChR and CD163(P<0.05,P<0.01),and decrease the fluorescence intensity of CD86(P<0.05,P<0.01).CONCLUSION Tianfu Zhixuan Formula may regulate the polarization of PVN microglia,reduce neuroinflammation and lower blood pressure in SHR rats by promoting α7nAChR expression.
