祛风除痹成膜凝胶制备及其抗炎镇痛作用、安全性评价OA
Preparation of film-forming gel for expelling wind and removing paralysis and evaluation of its anti-inflammatory and analgesic effects,safety
目的 制备祛风除痹成膜凝胶,并评价其抗炎镇痛作用和安全性.方法 以乌头茎叶为原料,制备成膜凝胶.在单因素试验基础上,以成膜剂(PVA1788)用量、增塑剂(丙三醇)用量、无水乙醇用量为影响因素,成膜时间、外观性状的综合评分为评价指标,正交试验优化制备工艺.采用醋酸致小鼠扭体模型考察镇痛作用,佐剂诱导大鼠关节炎模型考察抗炎作用,家兔皮肤刺激性实验考察安全性.结果 最佳条件为基质(MC-400)用量 0.3 g,成膜剂用量 0.1 g,增塑剂用量 0.5 g,无水乙醇用量 1 g,药物用量 0.5 g,综合评分为 33.69 分.与对照组比较,祛风除痹成膜凝胶中、高剂量组小鼠扭体次数减少(P<0.01),其中高剂量组疼痛抑制率为 50.94%;与模型组比较,祛风除痹成膜凝胶各剂量组大鼠足趾肿胀度、关节炎指数、血清 TNF-α 水平降低(P<0.05,P<0.01),并且中、高剂量组血清 IL-6、IL-1β 水平降低(P<0.05,P<0.01).末次给药后 1、24、48、72 h,家兔完整、破损皮肤未出现异常,无刺激性.结论 该方法稳定可靠,可用于制备抗炎镇痛较强、安全性良好的祛风除痹成膜凝胶.
AIM To prepare the film-forming gel for expelling wind and removing paralysis,and to evaluate its anti-inflammatory and analgesic effects,safety.METHODS The film-forming gel was prepared with stems and leaves of Aconitum carmichaelii Debx.as raw material.With film-forming agent(PVA1788)consumption,plasticizer(glycerol)consumption and anhydrous ethanol consumption as influencing factors,the comprehensive score for film-forming time and appearance characters as an evaluation index,the preparation process was optimized by orthogonal test on the basis of single factor test.Acetic acid-induced mouse writhing model,adjuvant-induced rat arthritis model and rabbit skin irritation test were employed to investigate anti-inflammatory effect,analgesic effect and safety,respectively.RESULTS The optimal conditions were determined to be 0.3 g for substrate(MC-400)consumption,0.1 g for film-forming agent consumption,0.5 g for plasticizer consumption,1 g for anhydrous ethanol consumption,and 0.5 g for drug consumption,and the comprehensive score was 33.69.Compared with the control group,the film-forming gel for expelling wind and removing paralysis medium-dose and high-dose groups demonstrated reduced mouse writhing frequency(P<0.01),among with the high-dose group displayed the pain inhibitory rate of 50.94%.Compared with the model group,the film-forming gel for expelling wind and removing paralysis various dose groups demonstrated decreased rat paw swelling degree,arthritis index and serum TNF-α level(P<0.05,P<0.01),and decreased serum IL-6,IL-1β levels were observed in the medium-dose and high-dose groups(P<0.05,P<0.01).At 1,24,48,72 h after last administration,the abnormalities in rabbit unbroken,broken skins did not occur,exhibiting non-irritation.CONCLUSION This stable and reliable method can be used for the preparation of film-forming gel for expelling wind and removing paralysis with strong anti-inflammatory and analgesic effects and good safety.
