基于HPLC指纹图谱、化学模式识别和含量测定评价黄花倒水莲质量OA
Quality evaluation of Polygala fallax based on HPLC fingerprints,chemical pattern recognition and content determination
目的 评价黄花倒水莲质量.方法 建立 HPLC 指纹图谱,分析采用 Waters CORTECS T3 C18色谱柱(2.1 mm×150 mm,1.6 μm);流动相乙腈-0.1%甲酸,梯度洗脱;体积流量 0.3 mL/min;柱温 35℃;检测波长 256 nm.再进行聚类分析、主成分分析、正交偏最小二乘法-判别分析,并以 4-β-O-葡萄糖基-1,3,7-三羟基呫吨酮为内标,计算7-O-甲基杧果苷、远志(口山)酮Ⅲ相对校正因子,一测多评法测定其含量.结果 29 批药材指纹图谱中有 5 个共有峰,相似度(除 S2 外)均大于 0.80.各批药材聚为 2 类,7-O-甲基杧果苷为质量差异标志物.3 种成分在各自范围内线性关系良好(R2≥0.999 0),平均加样回收率 95.34%~104.64%,RSD 1.31%~2.30%.结论 该方法准确可靠,可为黄花倒水莲质量控制提供依据.
AIM To evaluate the quality of Polygala fallax Hemsl.METHODS The HPLC fingerprints were established,the analysis was performed on a 35℃thermostatic Waters CORTECS T3 C18 column(2.1 mm×150 mm,1.6 μm),with the mobile phase comprising of acetonitrile-0.1%formic acid flowing at 0.3 mL/min in a gradient elution manner,and the detection wavelength was set at 256 nm.Subsequently,cluster analysis,principal component analysis and orthogonal partial least squares discriminant analysis were performed,after which lancerin was used as an internal standard to calculate the relative correction factors of 7-O-methylmangiferin and polygalaxanthone Ⅲ,and the content determination was made by quantitative analysis of multi-components by single-marker(QAMS)method.RESULTS There were 5 common peaks in the fingerprints for 29 batches of medicinal materials with the similarities of more than 0.80(except for S2).Various batches of medicinal materials were clustered into two categories,7-O-methylmangiferin was quality differential marker.Three constituents showed good linear relationships within their own ranges(R2≥0.999 0),whose average recoveries were 95.34%-104.64%with the RSDs of 1.31%-2.30%.CONCLUSION This accurate and reliable method can provide evidence for the quality control of P.fallax.
