目的 设计并合成一种高尔基体(golgi apparatus,GA)靶向近红外荧光探针MB-GA,探究其对次氯酸(hypo-chlorous acid,HClO)的光谱响应特性及活细胞成像性能,为亚细胞水平实时监测GA内HClO动态变化提供潜在工具.方法 以亚甲蓝(methylene blue,MB)为近红外荧光母核、4-(2-氨乙基)苯磺酰胺为GA靶向单元、内酰胺为HClO响应开关,设计并合成探针MB-GA.通过核磁共振氢谱/碳谱、高分辨质谱及密度泛函理论计算进行结构验证.利用紫外-可见和荧光分光光度计分析其光谱响应性能,CCK-8法检测其对HepG2和4T1细胞增殖活性的影响,激光共聚焦成像验证GA靶向定位能力及其对外源/内源HClO的成像效果.结果 MB-GA合成简便,产率达65%.MB-GA的HOMO-LUMO能隙高于MB(4.0 864 eV vs 2.4 800 eV),且振子强度较弱(f=0.0 010),摩尔消光系数(3.0×103 L⋅mol-1⋅cm-1)偏低.MB-GA对HClO响应迅速(约10 s)、灵敏度高(检测限0.067 μmol/L)、选择性好,且在pH为4.0~11.0和连续光照下光稳定性良好.MB-GA 对 HepG2 和 4T1 细胞增殖的抑制作用呈剂量依赖性(均 P<0.001),其中 MB-GA 浓度为10 μmol/L时,细胞的存活率高于50.0%.共聚焦成像显示,MB-GA具有一定的GA靶向能力(皮尔逊相关系数0.6);加入外源HClO或LPS诱导后荧光显著增强,且在肿瘤细胞HepG2(50.67±2.08)、4T1(59.33±3.65)细胞的荧光增强幅度明显大于正常肝细胞THLE-3(26.33±2.52).结论 本研究成功构建了一种可靶向GA的HClO响应型荧光探针MB-GA,该探针光谱性能良好并能有效区分癌细胞与正常细胞,有良好的潜在应用价值.

Objective To design and synthesize a novel Golgi apparatus(GA)-targeted near-infrared fluorescent probe,MB-GA,and to investigate its spectral response to hypochlorous acid(HCIO)and its cellular imaging capability,thereby providing a reliable tool for monitoring dynamic changes in HCIO within the GA at the subcellular level.Methods MB-GA was designed and synthesized using methylene blue(MB)as the near-infrared fluorophore,4-(2-aminoethyl)benzenesulfonamide as the GA-targeting moiety,and a lactam group as HCIO responsive switch.The structure of the probe was verified by ¹H NMR and ¹³C NMR spectroscopy,high-resolution mass spectrometry,and density functional theory(DFT)calculations.The spectral response properties of MB-GA were evaluated using ultraviolet-visible absorption spectroscopy and fluorescence spectrophotometry.CCK-8 assay was used to assess the viability of HepG2 and 4T1 cells.Confocal laser scanning microscopy imaging was further applied to verify the GA-targeting ability of MB-GA,as well as its imaging performance in response to exogenous and endogenous HCIO.Results MB-GA features a simple synthetic route with a total yield of 65%.DFT calculations showed that the HOMO-LUMO energy gap of MB-GA was larger than that of MB(4.08 eV vs 2.47 eV),accompanied by a weak oscillator strength(f=0.0 010)and a low molar extinction coefficient(3.0×10³ L·mol⁻¹·cm⁻¹).MB-GA exhibited a rapid response to HCIO(approxi-mately 10 s),with high sensitivity(detection limit of 0.067 μmol/L),excellent selectivity,and good stability over a wide pH range of 4.0-11.0 and good photostability under continuous irradiation.The proliferation-inhibitory effect of MB-GA on HepG2 and 4T1 cells viability was dose-dependent(all P<0.001),and cell viability remained above 50.0%at 10 μmol/L.Confocal imaging showed that MB-GA showed GA-targeting ability,with a Pearson correlation coefficient of 0.6;fluorescence intensity increased significantly after addition of exogenous HCIO or lipopolysaccharide induction,and the fluorescence enhancement in HepG2(50.67±2.08)and 4T1(59.33±3.65)cancer cells was notably higher than that in normal hepatic THLE-3 cells(26.33±2.52).Conclusions This study successfully developed a GA-targeted HClO-responsive fluorescent probe MB-GA.The probe exhibits excellent spectral performance and enables discrimination of HClO-related fluorescence responses between cancer cells and normal cells,suggesting its potential for practical biological applications.
