粒用型和菜用型大豆鲜籽粒的比较转录组学分析OA

本研究利用转录组测序分析了粒用型和菜用型大豆鲜籽粒的基因表达差异,为大豆鲜籽粒品质形成机理提供理论依据和技术支撑.采用高通量测序技术(IlluminaHiSeq 4000)对粒用型大豆品种福豆234和菜用型大豆品种闽豆7号的R6期鲜籽粒进行转录组测序.共获得39.92 Gb序列信息,56 956个注释基因,其中注释到的新基因(new genes)和差异表达基因(differentially expressed genes,DEGs)分别为932个和2088个.以福豆 234为参考,闽豆7号中有918个DEGs上调表达,GO(gene ontology)分析涉及生物学过程中的谷氨酰胺代谢过程、(1->3)-β-D-葡聚糖生物合成过程、碳水化合物代谢过程等通路相关基因被显著富集;KEGG(kyoto encyclopedia of genes and ge-nomes)通路中半乳糖代谢、木质素、亚木质素和蜡生物合成、淀粉和蔗糖代谢等通路相关基因被显著富集;闽豆7号中有1170个基因下调表达,GO分析涉及生物学过程中的内质网未折叠蛋白反应、木葡聚糖代谢过程、细胞壁生物发生等通路相关基因被显著富集;KEGG通路中ABC转运蛋白、GPI锚定蛋白生物合成、内质网中的蛋白质加工等通路相关基因被显著富集.与淀粉和蔗糖代谢途径相关的48个DEGs中,有25个在粒用型大豆福豆234鲜籽粒中上调表达,23个在菜用型大豆闽豆7号鲜籽粒中上调表达.同时也利用qRT-PCR(quantitative real time polymerase chain reaction)技术验证了转录组测序的检测结果的可靠性.本研究将为大豆鲜籽粒品质改良提供理论依据.

In this study,we analyzed the gene expression differences between fresh seeds of grain and vegeta-ble soybean to providing theoretical basis and technical support for the mechanism of soybean fresh seed quality for-mation by transcriptome sequencing.High throughput sequencing technology(IlluminaHiSeq 4000)was used to per-form transcriptome sequencing on R6 stage fresh seeds of the grain soybean variety Fudou 234 and the vegetable soybean variety Mindou 7.A total of 39.92 Gb sequencing data were obtained,representing 56,956 annotated genes.Among them,932 new genes and 2088 differentially expressed genes(DEGs)were annotated.Taking Fudou 234 as a reference,918 DEGs were upregulated in Mindou 7.Among 918 upregulated genes,GO analysis revealed that genes related to biological processes such as glutamine metabolism,(1->3)-β-D-glucan biosynthesis,and carbohy-drate metabolism were significantly enriched;Genes related to galactose metabolism,lignin,lignin and wax biosyn-thesis,starch and sucrose metabolism,and other pathways were significantly enriched in the KEGG pathway.Simi-larly,1170 genes were downregulated in Mindou 7,and GO analysis showed significant enrichment of genes related to biological processes such as endoplasmic reticulum unfolded protein response,xyloglucan metabolism,and cell wall biogenesis in biological processes;Genes related to pathways such as ABC transporters,GPI anchored protein biosynthesis,and protein processing in the endoplasmic reticulum were significantly enriched in the KEGG path-way.Among the 48 DEGs related to starch and sucrose metabolism,25 were upregulated in the fresh seeds of grain soybean Fudou 234 and 23 were upregulated in the fresh seeds of fresh soybean Min Dou 7.At the same time,qRT-PCR technology was used to verify the reliability of transcriptome sequencing.This study will provide a theoretical basis for improving the quality of soybean fresh seeds.
