盐酸氯苯胍(ROBH)是化学合成类抗球虫药,因其药效良好且生产成本较低而广泛应用于养殖业.目前,关于ROBH潜在遗传毒性的研究资料有限,且存在较大争议.为明确ROBH的遗传毒性,本试验采用Ames试验、小鼠骨髓细胞微核试验和单细胞凝胶电泳试验,分别在基因、染色体和DNA水平研究ROBH的遗传毒性.结果显示,与灭菌蒸馏水组相比,4株试验菌株ROBH低剂量组(200、40、8 μg/皿)的平均回复突变菌落数均无显著差异(P>0.05),4株试验菌株ROBH高剂量组(5 000、1 000 μg/皿)的平均回复突变菌落数均显著降低(P<0.05).与阴性对照组相比,195.00和97.50 mg/(kg·bw)ROBH剂量组嗜多染红细胞(PCE)微核率均极显著升高(P<0.01).20和25 μmol/L ROBH剂量组人肝癌细胞(HepG2)均有明显的彗星拖尾现象;与对照组相比,20和25 μmol/L ROBH剂量组HepG2细胞尾部DNA百分率均极显著升高(P<0.01).结果表明,低剂量组ROBH对试验菌株无致突变性,高剂量组ROBH对试验菌株有细菌毒性.ROBH可诱发哺乳动物细胞染色体损伤和DNA断裂,表现出明确的遗传毒性,为ROBH安全性再评价提供了重要依据.

Robenidine hydrochloride(ROBH)is a chemically synthesized anticoccidial drug widely used in animal husbandry because of its good efficacy and relatively low production cost.At present,information on the potential genotoxicity of ROBH is limited and remains highly controversial.To clarify the genotoxicity of ROBH,the Ames test,mouse bone marrow cell micronucleus test,and single-cell gel electrophoresis assay were employed to investigate its genotoxic effects at the gene,chromosome,and DNA levels,respectively.The results showed that,compared with the sterile distilled water group,the average numbers of mutant colonies in the low-dose ROBH groups(200,40,and 8 μg/plate)of the four test strains were not significantly different(P>0.05),whereas the average numbers of mutant colonies in the high-dose ROBH groups(5 000 and 1 000 μg/plate)were significantly decreased(P<0.05).Compared with the Negative control group,the micronucleus rates of polychromatic erythrocytes(PCEs)in the 195.00 and 97.50 mg/(kg·bw)ROBH dose groups were significantly increased(P<0.01).Obvious comet tailing was observed in hepatocellular carcinoma cell line 2(HepG2)cells treated with 20 and 25 μmol/L ROBH.Compared with the Control group,the percentages of tail DNA in HepG2 cells in the 20 and 25 μmol/L ROBH groups were significantly increased(P<0.01).These results indicate that low-dose ROBH is non-mutagenic to the test strains,whereas high-dose ROBH shows bacterial toxicity to the test strains.ROBH can induce chromosomal damage and DNA strand breaks in mammalian cells,demonstrating clear genotoxicity.The findings of this study provide important evidence for the re-evaluation of the safety of ROBH.
