Purification process and antibacterial activity of total flavonoids from the aerial part of Atractylodes lancea

本试验以茅苍术地上部分为材料,采用超声法提取总黄酮,通过静态吸附-解吸附试验筛选最佳树脂型号;通过动态试验,以吸附率和解吸附率为指标,考察上样液浓度、上样速度、上样液体积以及洗脱剂浓度、洗脱速度和洗脱剂体积对大孔树脂纯化茅苍术地上部分总黄酮的影响,并通过正交法优化总黄酮纯化工艺;此外,通过平板法研究总黄酮对金黄色葡萄球菌和大肠杆菌的抑菌活性.结果显示:D101 为最适宜的树脂型号,最佳纯化工艺确定为:上样液浓度 30 mg/mL,上样速度1 BV/h,上样体积30 mL,使用60 mL 80%乙醇作为洗脱剂,控制洗脱速度为1 BV/h.使用该条件纯化后,茅苍术地上部分总黄酮的纯度可达到 48.2%.纯化后的总黄酮对金黄色葡萄球菌具有明显的抑菌作用,其最小抑菌浓度为2.26 mg/mL.

This experiment used the aerial part of Atractylodes lancea as the material,total flavonoids were extracted by ultrasonic method.Then the optimal resin model was selected based on the results of static adsorption and static desorption experiments;Using adsorption rate and desorption rate as indicators,the effects of sample concentration,sample loading speed,sample loading volume,and eluent concentration,elution speed,and eluent volume on the purification process of total flavonoids were investigated,and the total flavonoid purification process was optimized through orthogonal design.In addition,the antibacterial activities of total flavonoids against Staphylococcus aureus and Escherichia coli were studied using the agar plate method.The results showed that D101 macro-porous resin was the most suitable one.The purification process involved a sample concentration of 1.20 mg/mL,a sample loading rate of 1 BV/h,a sample volume of 30 mL,and 60 mL of 80%ethanol as the eluent.The elution rate was controlled at 1 BV/h.Using this purification condition,the total flavonoid purity could reach 48.2%.The total flavonoids obtained had a significant antibacterial effect on Staphylococcus aureus,with a minimum inhibitory concentration of 2.26 mg/mL.
