基于镜检与DNA宏条形码技术的恰甫其海伊犁鲈食性特征研究OA
Study on dietary characteristics of Perca schrenkii in Qiapuqihai Reservoir based on microscopy and DNA metabarcoding
伊犁鲈(Perca schrenkii)作为新疆伊犁河支流水库的优势经济鱼类,研究伊犁鲈的食性有助于明确其摄食对象与营养级位置.本研究采用镜检分析与DNA宏条形码技术相结合的方法,系统分析新疆恰甫其海(Qiapuqihai Reservoir)伊犁鲈的摄食强度、食物组成及季节性变化.结果表明,伊犁鲈全年摄食率高达 93%,秋季摄食强度最高,其次为春季,夏季摄食强度最低.镜检结果显示,伊犁鲈食物组成以浮游动物[溞属(Daphnia)、华哲水蚤属(Sinocalanus)]和底栖动物[摇蚊属(Chironomus)]为主,鱼类[棒花鱼(Abbottina rivularis)、银鲫(Carassius auratus gibelio)]占比次之.镜检法鉴定出小型鱼类等饵料,DNA宏条形码技术额外鉴定出 8属未被镜检识别的微型生物,两种方法在属级水平的重合率为 51.5%,互补性显著.对不同季节伊犁鲈饵料生物进行相似性分析(ANOSIM),结果表明不同季节饵料组成差异显著(P<0.05);非度量多维尺度分析(nMDS)结果显示,季节间样本分布可区分,春季饵料种类较丰富,夏季相对单一.研究表明,两种方法结合显著提升了饵料鉴定的完整性,进而系统揭示了伊犁鲈的摄食生态特征.相关研究结果可为恰甫其海水域生态系统能量流动解析及渔业资源管理提供理论依据.
As an economically dominant fish species in tributaries and reservoirs of the Ili River Basin in Xinjiang,studying the feeding habits of Perca schrenkii is essential to clarify its prey and trophic level.We used a combination of microscopic examination and DNA metabarcoding to systematically analyze the feeding intensity,dietary composition,and seasonal variations of Perca schrenkii in Qiapuqihai Reservoir,Xinjiang.The results indicated that the annual feeding rate of Perca schrenkii reached 93%,with the highest feeding intensity observed in autumn,followed by spring;the lowest was observed in summer.Microscopic examination revealed that the diet primarily consisted of zooplankton(Daphnia and Sinocalanus)and benthic organisms(Chironomus),followed by fish(Abbottina rivularis and Carassius auratus gibelio).Microscopy complemented by DNA metabarcoding identified prey such as small fish and revealed eight additional genera of microorganisms,resulting in a 51.5%genus-level overlap and demonstrating significant methodological complementarity.Similarity analysis of prey organisms across seasons revealed significant differences in dietary composition(P<0.05).Non-metric multidimensional scaling results indicated distinguishable distributions of samples among seasons,with a richer prey diversity in spring and a relatively simpler composition in summer.The results of this study demonstrate that combining both methods significantly enhances prey identification comprehensiveness,thereby systematically revealing the ecological feeding characteristics of Perca schrenkii.These findings provide a theoretical foundation for understanding energy flow in the Qiapuqihai Reservoir aquatic ecosystem and managing fishery resources.
