栉孔扇贝LAMP1基因的序列特征、表达模式及功能OA
Sequence characteristics,expression,and function of the LAMP1 gene in Chlamys farreri
基于实验室前期组学筛选,推测LAMP1基因与栉孔扇贝精巢发育相关.本研究利用生物信息学方法分析了栉孔扇贝(Chlamys farreri)LAMP1基因的序列特征,利用荧光定量PCR检测了LAMP1基因在增殖期栉孔扇贝肾脏、肝胰腺、闭壳肌、鳃、外套膜及性腺等各组织中的表达模式,及其在不同性腺周期栉孔扇贝性腺中的表达模式,并利用RNA干扰技术探究了LAMP1基因的功能.结果显示:栉孔扇贝LAMP1 CDS长 1926 bp,编码 641个氨基酸,包含高度糖基化的管腔结构域、独特铰链区、C-末端的跨膜保守区以及短的胞质尾部.该基因在栉孔扇贝雌雄各组织的表达有差异,表现为除外套膜外,其他组织均为雄性表达高于雌性,特别是精巢中的表达量达卵巢表达量的 3.27倍;雄性组织中精巢表达量最高,肾脏中也有较高表达;雌性组织中外套膜和卵巢表达量较高.体外合成LAMP1基因dsRNA后注入扇贝闭壳肌中(第一次注射 5 d后进行第二次注射),第 12 d取样检测发现,实验组扇贝精巢(卵巢)中的LAMP1 mRNA表达量相对于空白对照组降低了 65%(62%),较阴性对照组降低 62%(60%),目标基因被有效敲降;干扰后的卵巢无明显变化,但精巢中,精子形态出现异常,长度短于对照组,形状改变为圆球型.综合认为,LAMP1主要在栉孔扇贝精巢发挥作用,影响扇贝精细胞形成精子的过程,推测该基因通过参与自噬清除结构异常的精子,维持精子的正常形态.
Lysosome-associated membrane protein 1(LAMP1)is a member of the LAMP family;current studies on this gene have mainly focused on vertebrates,wherein it is considered to be involved in spermatogenesis.To date,no studies have examined LAMP1 in mollusks.Based on preliminary omics analyses conducted in our laboratory,LAMP1 is presumed to be associated with testis development in Chlamys farreri.Therefore,a detailed investigation of this gene was conducted to provide support for a better understanding of testis development in C.farreri.Bioinformatic approaches were used to analyze the sequence characteristics of LAMP1 in C.farreri.Quantitative reverse transcription PCR(qRT-PCR)was employed to examine the expression patterns of LAMP1 in various tissues,including the kidney,hepatopancreas,adductor muscle,gill,mantle,and gonad,as well as across different stages of the gonadal cycle.Additionally,RNA interference(RNAi)was used to investigate the functional role of LAMP1.Bioinformatic analysis revealed that LAMP1 had a coding sequence of 1926 bp,encoding 641 amino acids.The protein contained a highly glycosylated luminal domain,a unique hinge region,a conserved C-terminal transmembrane region,and a short cytoplasmic tail.qRT-PCR revealed differences in LAMP1 expression between sexes and among tissues.In most tissues,male scallops had higher LAMP1 expression than female ones,except in the mantle.Testis expression was 3.27 times greater than ovary expression.In males,the testis exhibited the highest expression,followed by the kidney.In females,the mantle and ovary exhibited relatively high expression levels.RNAi successfully reduced LAMP1 mRNA expression in the experimental group by 65%in ovaries and 62%in testes compared with the blank control and by 62%in ovaries and 60%in testes compared with the negative control.Morphological analysis after RNAi treatment revealed no significant changes in the ovary;however,the testis displayed abnormal sperm morphology.The sperm were short and spherical,unlike the typical elongated shape seen in the controls.LAMP1 is predominantly expressed in the testes of comb-toothed scallops.It is mainly involved in the differentiation of spermatogonia into spermatozoa.This gene is hypothesized to regulate normal sperm morphology.It may facilitate the autophagic clearance of structurally abnormal spermatozoa.
